Objective:The Endoglin belonging angiogenesis inhibitor, are mainly expressed on the surface of endothelial cells and placental syncytiotrophoblast cell.They are essential markers about proliferation of endothelial cell and angiogenesis.By inhibiting TGF-β signal transduction,the Endoglin plays a biologicallearning effect and critical negative regulatory role in the process of regulating the differentiationtrophoblast cells. Domestic and international study found that the over expression of Endoglin in patients with hypertensive disorders complicating pregnancy(HDCP),and speculated Endoglin can inhibit the growth and migration of trophoblast cells, bring about placental vascular remodeling obstacles and shallow implantation of the placenta, cause placental blood flow perfusion decrease. Liver X receptor (liver X receptor, LXR) is a member of the nuclear receptor superfamily, including LXRa and LXRβ1in mammals.LXRs as an oxidized steroids receptors involved in regulating the body a variety of physiological activities, including the process of metabolism and transporter of cholesterol, fat formation, gluconeogenesis and inflammation. LXRa expressed in trophoblast and decidual cells involved in regulating placental lipid metabolism. Chinese and foreign scholars found that LXRa expression increased in serum and placenta in patients with HDCP, especially in preeclampsia,suggested that LXRs play an important role in the development of hypertensive disorders. There are few reports at home and abroad about the pathogenesis and correlation of Eng and LXRa of hypertensive disorders in pregnancy.This study examines the expression of Eng gene and LXRa gene in the placentas of hypertensive disorders in pregnancy through fluorescence quantitative real-time PCR, in order to explore changes of Eng and LXRa gene in placenta of HDCP and their clinical significance.Methods:From Aug2010to Apr2011,44patients with hypertensive disorderscomplicating pregnancy (HDCP) who delivered in General Hospital of Beijing Military Command were recruited in the study group(HDCP group), including15women with gestational hypertension(GH group),14women with moderate preeclampsia(MPE Group) and15women with severe preeclampsia (SPE group). Another16healthy pregnant women were taken as control group. Quantitative Real Time RT-PCR was used to investigate the expression of Endoglin nd LXRa mRNA in placenta.Results:1. the expression of Endoglin mRNA in the placenta of HDCP group were significantly elevated than that in control group(P<0.05).2. The expression of Endoglin mRNA in placenta of MPE group were higher than that of control group(P<0.05).3. The expression of Endoglin mRNA in placenta of SPE group were higher than that of control group(P<0.05).4.The expression of LXRa mRNA in the placenta of HDCP group were significantly elevated than that in control group(P<0.05);5.The expression of LXRa mRNA in placenta of MPE group were obviously higher than that of control group(P<0.05).6. The expression of LXRa mRNA in placenta of SPE group were significantly elevated than that in MPE group(P<0.05).7.There was a significant positive relationship between Endoglin and LXRa in placenta in HDCP group.Conclusion:1. The Endoglin and LXRa gene both express in placenta of healthy pregnant women and patients with HDCP, but the expression quantity was difference2. The over expression of Endoglin and LXRa gene in the placenta may be closely related with the occurrence of HDCP and the expression quantity related to the severity of the disease.3. There was a correlation of between Endoglin and LXRa in placenta in HDCP,but the interaction in HDCP pathogeresis remain to be further reseach.4. Further explore the relationship of Endoglin and LXRa gene can provide a new way of prevention and treatment of HDCP.
