Objective:To investigate the gut mucosal barrier function effect of the Chinese medicine Rhubarb inrats with gastrointestinal surgery.Methods:Thirty male Sprague Dawley (SD) rats were randomly divided into three groups:Control(C),enteral nutrition(EN)group, the daihuang-enriched enteral nutrition (D-EN) group.EN Group and G-EN Group received Cecectomy and gastrostomy with the use of50mgamoxicillin and20mg metronidazole twice.Each group would be treated with isocaloric andisonitrogenous nutrition for next7days after the operation, respectively. Taking5cm terminalileum and1cm intestines, the morphology of intestinal epithelial tight junctions can be observedby optical microscopy. For each group, then solid-phase sandwich enzyme-linkedimmunosorbent assay (ELISA) was used to compare levels of plasma D-lactate, and testintestinal mucosal permeability.The transmembrane binding proteins (occludin) were measuredwith immunohistochemistry.Results:For EN Group, intestinal mucosa obviously atrophied and the villus height, crypt depth,mucosal thickness, villus surface area of ileum and transmembrane binding proteins (occludin)were lower than those of rats of C Croup, and D-EN Group (P<0.05), but D-lactate were muchhigher than other Groups’(P<0.05). There was no significant difference between D-EN Groupand C Group in the ileal membrane morphology transmembrane binding proteins (occludin)expression level, intestinal mucosa permeability and level of TNF-α(P>0.05).Conclusion:Chinese medicine Rhubarb can significantly improve the gut mucosal barrier function effect of the rat with gastrointestinal surgery, can effectively maintain intestinal epithelial morphology,structure and barrier function, increase intestinal epithelial occludin protein expression, thusspeeding up the intestinal barrier function after gastrointestinal surgeryrecovery.
