Objective To investigate the expression and clinical implications of MACC1andc-Met in gastric cancer.Methods Collected gastric carcinoma and adjacent normal tissues.The expression ofMACC1mRNA and protein in three gastric cancer lines, gastric carcinoma and adjacentnormal tissues in46cases were detected by real-time fluorescence quantitative polymerasechain reaction (RT-qPCR).MACC1and c-Met protein in gastric carcinoma and adjacentnormal tissues in67cases were detected by immunohistochemistry method respectively. Therelationship between MACC1, c-Met protein expression and the clinicopathological featuresof patients with gastric cancer were analyzed.Results MACC1mRNA differentially expressed in gastric cancer cell lines.Theexpression levels of MACC1mRNA were higher in gastric carcinoma(5.5600±2.8531)thanthose in adjacent normal tissues(2.1901±1.6759)(t=6.907,P<0.05).The positive rates ofMACC1protein in gastric cancer tissues(64.18%,43/67)were significantly higher than that inthe adjacent normal tissues (5.97%,4/67),(χ~2=49.844,P<0.05).The expression of MACC1protein in the gastric cancer tissue was evidently correlated with the degree of histologicaldifferentiation(χ~2=14.492,P=0.000),the peritoneal metastasis(χ~2=4.251,P=0.039),the lymphnode metastasis(χ~2=11.978,P=0.001) and the clinical stage(χ~2=11.024,P=0.001), but not withage, gender, tumor size, location or distant metastasis(P>0.05).The positive rates of c-Metprotein in gastric cancer tissues(65.67%,44/67)were significantly higher than that in theadjacent normal tissues(8.96%,6/67),(x2=46.070,P<0.05).The expression of c-Met protein in the gastric cancer tissue was evidently correlated with the degree of histologicaldifferentiation(x~2=6.160,P=0.013),the peritoneal metastasis(x~2=6.560,P=0.010),the lymphnode metastasis(x~2=6.063,P=0.014) and the clinical stage(x~2=5.939,P=0.015),but not with age,gender, tumor size, location or distant metastasis(P>0.05).Conclusion The positive rate of MACC1protein expression was associated with c-Metprotein expression. They are regarded as predictors of peritoneal metastasis, lymph nodemetastasis and effective indicators of the evaluation of gastric cancer progress, which canprovide the basis for clinical diagnosis and treatment of gastric cancer patients.
