[Objective]To research influence of Pygo2in human renal cell carcinoma OS-RC-2cell lines. To provide tools for the study of the mechanism of human cell carcinoma and the targeted therapy.[Methods]Use human renal cell carcinoma cancer OS-RC-2cell lines and after a series of different treatments of Pygo2, then grouped transplanted into nude mice model tumorigenicity through Pygo2of kidney cancer pathogenesis in vivo. MTT method was used to detect the proliferation of OS-RC-2cell.16mice divided into four groups, each group of four, respectively, with the four treated renal cancer cell suspensions, Group A:blank control group of experimental control group GFP (original cells), B group Pygo2scrRNA (blank vector implant), C group of the expression group Pygo2OE, D group silent group Pygo2shRNA (using gene knockout technology to make Pygo2low expression),4.0×106renal cell carcinoma cell suspension0.2ml planted in nude front right armpit. Then the formation of transplanted tumors was studied. IHC detected that each tumor invasive protein (MMP-9, MMP-7and VEGF).[Result]2days cells into the logarithmic growth phase. A, B, C tumor rates were100%, D composed of tumor was75%(experiment was repeated twice). Pygo2OE group bean size transplanted tumors visible in three days,7days, the GFP and Pygo2scrRNA group touched subcutaneous the Summary, Pygo2shRNA10transplanted tumors. Group C tumor formation rate was significantly better than the other three groups (P <0.05). Group D of the speed of tumor formation and the tumor were lower than in the other groups (P<0.05).IHC analysis revealed that pygo2shRNA reduced the levels of matrix metalloproteinase-7(MMP-7) and VEGF in the OS-RC-2cells in vivo, however, the loss of Pygo2can not obviously decreased the expression of MMP-9, compared with reduction of MMP-7and VEGF expression quantity phase.[Conclusion]Using OS-RC-2cell lines, after its Pygao2intervention successfully established in nude mice model. Different packet tumor into the tumor volume differences, which provide a theoretical basis for in vivo studies of kidney cancer targeted therapy, has important significance.
