Objective To examine the expression change of HIF-1α and VEGF in gene levelsin MUTZ-1cell line under chemical hypoxia induced by Cobalt chloride in vitro,which explored the role in myelodysplastic syndrome angiogenesis.To investigate theeffect of different concentrations of thalidomide on the expression change of HIF-1αand VEGF in gene levels in MUTZ-1cell lin under hypoxic environment,whichexplored its possible mechanism in the treatment of myelodysplastic syndrome.Methods (1) Different concentration of Cobalt chloride was intervened for4hours、8hours、12hours and24hours, culture the MUTZ-1cell line invitro.Proliferation of MUTZ-1cells were tested by MTT analysis.(2) Aftercultured in hypoxia environment induced by Different concentration of Cobaltchloride for different times,expression of HIF-1α and VEGF gene were tested by realtime fluorescence quantitative reverse transcription-polymerase chain reaction.(3)Different concentration of thalidomide was intervened for24hours,culture theMUTZ-1cell line in vitro for hypoxia.Proliferation of MUTZ-1cells were tested byMTT analysis.(4) After cultured by Different concentration of thalidomide for24hoursfor hypoxia,expression of HIF-1α and VEGF gene were tested by real timefluorescence quantitative reverse transcription-polymerase chain reaction.Results (1) Different concentration of Cobalt chloride has different effects onMutz-1cell,50μmol/L Cobalt chloride has a little,with higher dose,Cobalt chloridecould further inhibit the proliferation of mutz-1cells.The difference between thesegroups had statistical significance(P<0.05).with higher time,cell viability has nosignificant changes.(2) Expression of HIF-1α and VEGF mRNA under hypoxiaenvironment induced by Different concentration of Cobalt chloride is higher than normoxia.The difference between treated group and untreated group had statisticalsignificance(P<0.05).In the same concentration of Cobalt chloride but differenttimes, the difference between these groups on expression of HIF-1α and VEGFmRNA had no statistical significance(P>0.05).100μmol/L Cobalt chloride group washigher than50μmol/L Cobalt chloride group and150μmol/L Cobalt chloride group onexpression of HIF-1α and VEGF mRNA.(3) Thalidomide could significantly inhibitthe proliferation of mutz-1cells for hypxia,with higher dose,cell viability hasGradually reduced, the difference between these groups had statisticalsignificance(P<0.05).(4) Different concentration of thalidomide treated groups waslower than untreated group on expression of HIF-1α and VEGF mRNA for hypoxia,with higher dose,expression has Gradually reduced, the difference between thesegroups had statistical significance(P<0.05).Conclusion (1)MUTZ-1cell growth was inhibited under the hypoxia environmentinduced by Cobalt chloride.(2)MUTZ-1cell could up-regulate the expression ofHIF-1α and VEGF gene under the hypoxia environment induced by Cobaltchloride.(3)After cultured by thalidomide for hypoxia,MUTZ-1cell growth wasinhibited.(4)After cultured by thalidomide for hypoxia,MUTZ-1cell coulddown-regulate the expression of HIF-1α and VEGF gene....
