Objective: To study the effect of cobalt pretreatment on 5/6 nephrectomized rats and discuss the mechanisms. Methods: Male Sprague-Dawley rats were subjected to 5/6 nephrectomy and randomly assigned to Cobalt-treatde group(Co),Remnant kidney group(RK), and sham-operated group(So). Serum creatinine, urea and 24h U-protein were measured in different time. Pathologic changes within kidney were investigated and immunohistochemistry were employed to investigate the expression of VEGF,HIF,TGF-β1,α-SMA. Results: Cobalt pretreatment reduce serum creatinine, urea and 24h U-protein. The deposition ofα-SMA,TGF-β1 was decreased and the HIF,VEGF expression was elevated.Conclusions: Cobalt pretreatment attenuate pathological lesions of residual kidney may through mechanisms such as induction of HIF-regulated genes,promotion of angiogenesis and conservation of capillary networks. |
