Objective To investigate the mechanism of Vimentin in the apoptosis of humanpancreatic cancer cell line induced by the TPL.Methods In vitro, study group was treated by TPL, control group was treated byPBS. Human pancreatic caner line AsPC-1was treated by TPL. We have used aproteomic approach to identify proteins that induce apoptosis in pancreatic caner cellline AsPC-1, The difference of the proteomics between the study group and controlgroup was investigated by the Two-dimensional PAGE, Protein identification usingelectrospray mass spectroscopy was done at the Fudan University Research Institute.The protein level of Vimentin was detected by Western blotting. Cell apoptosis ratewas detected by flow cytometry(FCM) with Annexin V/PI double stainings.Results Vimentitn isoforms in study group were observed by the Two-dimensionalPAGE comparing with the control group (P<0.01). After induced by TPL for0h,8h,24h,48h,The protein level of Vimentin detected by Western blot were graduallydecreasing (P<0.01). Then treated by TPL in the concentration of0ng/ml,12.5ng/ml,25ng/ml,50ng/ml,The protein level of Vimentin was alsodecreasing with the increasing concentration of the TPL (P<0.01). We also treated theAsPC-1with TPL,TPL plus caspase3,7-inhibitor and PBS, After8hours, theexpression of Vimentin in the combination group and the control group were morethan the single drug group (P <0.01).Conclusion The apoptosis of humane pancreatic caner cell line AsPC-1induced byTPL is relevant with the caspase cleavage of Vimentin.
