Objective The lack of effective treatment for pancreatic cancer results in a very lowsurvival rate.Gemcitabine is the first-line drug on the chemotherapy of pancreatic cancer,but the overall effect is poor.This study explores the enhancement of the therapeuticeffect on human pancreatic cancer cell line AsPC-1via the combination of gemcitabineand triptolide. Methods Proliferation inhibition of AsPC-1cells treated by GEM, TPL,and GEM combined with TPL was determined by MTT assay; The expression changes ofPARP and caspase-3were detected by Western Bloting; The differences of cell apoptosisratios were analyzed by flow cytometer. Results The proliferation of AsPC-1cellswas inhibited by TPL or GEM alone, with dose and time related.48h IC50of GEM onAsPC-1cells was720.67±5.67ng/ml,and72h IC50was309.48±3.35ng/ml;48hIC50of TPL on AsPC-1cells was14.23±1.45ng/ml,72h IC50was1.69±0.24ng/ml;TPL combined with GEM on AsPC-1cells, the two drugs had a certainsynergistic effect. After48hours, the differences of expression amount of activatedcaspase-3had statistically significant when combination group compared with othergroups (P <0.01); Single drug group compared with control group,with significantdifference (P <0.01). After48hours, the cleavaged PARP expression of combinationgroup compared with single drug group and control group, with significant difference (P<0.01); TPL single group and GEM single group compared with control group, withsignificant difference (P <0.01); After24hours, compared combination group, TPLsingle group,GEM single group with control group, the differences of cells apoptosisrates were statistically significant (P <0.01); Combination group compared with GEMalone group and TPL alone group,with significant difference (P <0.01). After48hours,the cells apoptosis rates of combination treatment group, TPL singl group,GEM singlgroup compared with control group, with significant difference (P <0.01); Combinationgroup compared with GEM alone group and TPL alone group, with significant difference(P <0.01).GEM and TPL two inhibition rate(0.5-1) when the two drugs combined with the cooperativity parameter (CI)valued of1.262-4.8944. Conclusions TPL hassignificant effect of inhibitory proliferation and inducing apoptosis in AsPC-1cells; TPLcombined with GEM shows certain synergistic effects on inhibiting proliferation ofAsPC-1cells. TPL could enhance GEM-induced apoptosis in AsPC-1cells.
