【Objective】To probe the effect of PNS in prevention of peritoneal fibrosisestablished by4.25%dextrose solution with lipopolysaccharide inrats.【Methods】Twenty-one Wistar rats were randomly divided into three groups:control group (n=7), received100ml· kg-1normal saline dailyintraperroneally (IP)for4weeks; model group(n=7): received100ml·kg-14.25%peritoneal dialysate daily IP for5weeks combined with fourinjections of lipopolysaccharide (0.6mg·kg-1on days1、3、5、7);experimental group(n=7):received100ml·kg-14.25%peritoneal dialysateplus35mg·kg-1PNS daily IP for4weeks combined with four injections oflipopolysaccharide (0.6mg·kg-1on days1、3、5、7).At the end of the studya l hour peritoneal equihbration testwas performed and Dialysis solution/plasma ureophil ratio (D/P), reuptake of glucose (D1/D0),ultrafiltration volume and white blood cells counting in dialysissolution were estimsted. TGF-β1、CTGF、MCP-1in the peritoneal dialysissolution was quantified by ELISA assay, and mRNA of TGF-β1、CTGF、MCP-1in the peritoneal tissue was quantified by RT-PCR. Structural variationsof peritoneal tissue were respectively observed by microscope.【Results】Compared with control rats, ultrafiltration volume in of the modelgroup and experimental group was significantly attenuated (P <0.05),while the total number of white blood cells and TGF-β1、CTGF、MCP-1in the dialysis solution, and TGFβ1mRNA、 CTGF mRNA、MCP-1mRNA in theperitoneal tissue and peritoneal thickness、 collagen deposition in theperitoneal tissue were significantly increased (P <0.05). Theultrafiltration volume in experimental group was significantly inmprovedthan in model group (P <0.05), nevertheless, the total number of whiteblood cells and TGF-β1、CTGF、MCP-1in the dialysis solution weresignificantly decreased, and TGFβ1mRNA、 CTGF mRNA、MCP-1mRNA in theperitoneal tissue did,both of peritoneal thickness and collagendeposition in the peritoneal tissue were obviously improved either (P <0.05).【Conclusion】1、A type of peritoneal fibrosis model established by4.25%dextrose solution with lipopolysaccharide in rats could be successfullyestablished.2、PNS could protect peritoneal fibrosis and amelioratethe functional and structural parameters of peritoneum.3、 PNS could ameliorate peritoneal fibrosis throughdown-regulation of TGFβ1、CTGF and MCP-1.
