Osteoporosis is a disease serious threat to human health, the light is broken, or life-threatening. The exact pathogenesis is not clear. In the world today there are three kinds of the most common cause of osteoporosis, namely postmenopausal osteoporosis and senile osteoporosis, drug-induced osteoporosis (glucocorticoids). This experiment adopts the ovariectomy rats to establish animal model of postmenopausal osteoporosis, glucocorticoid induced osteoporosis animal models,explore the effect of different dosage of GC on the bone mass and metabolism, and compare the different effect on the bone mass and metabolism between GC-induced osteoporosis modle and ovariectomy-induced osteoporosis.Methods:Total75,4-month-old female SD mice, were randomly divided into5group, including:A group, as the control, were treated with salt solution lavage; B group received the low dose GC lavage; C group were treated with moderate-dose lavage; D group were treated with high-dose GC lavage; E group receive ovariectomy.5mice were sacrificed to collect the femoral Metaphysis to process undecalcified bone histomorphometry and examine the lumbar1section by micro-CT, and testing the TRACP, G-ALP, BG of vein blood and DPD of urine by Elisa from all group at month4,8,12.Result:(1)after4weeks, the experimental group show that bone trabecular arrangement was gradually sparse, the figure has decreased, trabecular apparent necking, spacing widened.(2) the bone mineral density detection:compared with the control, bone loss of low dose group became obvious after the8weeks treatment (P<0.05), in medium dose and high dose group after4weeks (P<0.05). there are no significant difference in the each period of medium dose group and high dose group (P>0.05); compared to the4weeks castration group, the medium dose and high dose group have significant difference (P<0.05) at4week,but not at8and12weeks(P>0.05).(3) bone metabolism and biochemical index:and remove ovarian4weeks later, and normal control group. At4weeks post-surgery or GC lavage, compared to the control group, group C, D and E have significant difference (P<0.05) at bone formation markers (G-ALP, BGP), bone resorption marker (TRACP, DPD).Conclusion:(1) the bone mass lossing may along with the dose of glucocorticoids increase.(2) in OVX model, bone reconstruction ability was obviously increased. However, in the GIOP model this ability was drop.
