Objective The aim of this study was to determine the association between estrogenreceptors gene polymorphisms and chronic periodontitis in Hui and Han populations fromNingxia and to analyze the distributions of ER genotypes and alleles frequency between thetwo nationalities.Methods We recruited accordanced with the diagnostic criteria and inclusion criteria forthe study, including135patients with chronic periodontitis (CP) and139healthy controls inHan nationality;120patients with chronic periodontitis and126healthy controls in Huinationality. In our study, the genotypes of ER intron ER-α/XbaⅠ(rs9340799)andER-α/PvuⅡ(rs2234693)were investigated by polymerase chain reaction-restriction fragmentlength polymorphism (PCR-RFLP) respectively. We evaluated the genotypes and allelesfrequency of ER-α/XbaⅠ(rs9340799) and ER-α/PvuⅡ(rs2234693) from all the subjects andanalyzed that whether there are associations between ER gene polymorphisms and CP bycase-control methods.Results (1) ER-α/XbaⅠ(rs9340799): We found3genotypes(AA、AG and GG) ofrs9340799in all groups. The difference in the frequencies of ER-α/XbaⅠ(rs9340799)genotypes and alleles was no statistic significance between the CP group and HC group in Huipopulations(P=0.684and P=0.399, respectively); In Han populations, the difference in thefrequencies of genotypes and alleles was no statistic significance between the CP group andHC group(P=0.376and P=0.553, respectively). And the difference in the frequencies ofER-α/XbaⅠ(rs9340799)genotypes and alleles has no statistic significance between the Hui CP group and Han CP group(P=0.209and P=0.182, respectively). The difference in thefrequencies of genotypes and alleles also has no statistic significance between the Hui HCgroup and Han HC group (P=0.200and P=0.099, respectively).(2) ER-α/PvuⅡ(rs2234693):We found3genotypes (CC、CT and TT) of rs2234693in all groups. There was statisticallysignificant difference in the frequencies of ER-α/PvuⅡ(rs2234693) genotypes (P=0.036), nosignificant difference alleles (P=0.374) between the CP patients and healthy controls in Huinationality. In Han populations, the difference in the frequencies of genotypes and alleles wasno statistic significance between the CP group and HC group (P=0.492andP=0.873,respectively).But the difference in the frequencies of ER-α/PvuⅡ(rs2234693)genotypes and alleles has statistic significance between the Hui CP group and Han CPgroup(P=0.014and P=0.009, respectively); And the difference in the frequencies of genotypesand alleles has no statistic significance between the Hui HC group and Han HCgroup(P=0.282and P=0.109, respectively).Conclusion (1) The polymorphism of the ER-α//XbaⅠ(rs9340799) was not associatedwith the chronic periodontitis in Hui and Han nationality and there was no difference betweenthe two nationalities.(2) The polymorphism of the ER-α/PvuⅡ(rs2234693) was associatedwith the chronic periodontitis in Hui and Han nationality, the ER-α/PvuⅡ genotype may be asusceptible indicator for chronic periodontitis in Hui nationality.
