| Objective:1. To explore the method of isolation, culture, purification of human umbilicalcord-derived mesenchymal stem cells (hUCMSCs) in vitro and identification ofits biological characteristics for further study and clinic application.2. To establish an animal model for assessment on therapeuticeffect of hUCMSCs on firearm wound for further study.3. To propose a new therapy injecting with hUCMSCs after thedebridement of firearm wounds. We clarified the effect on firearm wounds inpigsMethod:1. To isolate the hUCMSCs from infant umbilical cores of healthy parturients bytissue explants adherent method. The shape was observed after subculture. Thecell membrane antigens were identified. The potency that hUCMSCs are inducedand differentiated into osteoblasts and lipoblasts was observed.2.8pigs were divided into group A and group B randomly. Hind limbs of animalsin group A and B were wounded respectively with projectiles fired by a56-typemusket and a53-type musket. Appearance, size of wound track entrance and exit,,maximum cross-sectional area of wound tracks, histopathologic zonation,bacterial content around the wound tracks were measured. Constancy ofmodel was evaluated.3.20Landrace wounded by a56-type musket were divided into group A and groupB randomly: wound incision+hUCMSCs (A), wound incision+sterilizingsaline (B).The effectiveness was estimated using bacterial content, histopathologic examination, and wound healing time.Result:1. CD90, CD29, CD73, CD105merebrane antigen could be detected of the thirdgeneration of hUCMSCs isolated by tissue explants adherent method, but notCD45, CD34, HLA-DR membrane antigen.The hUCMSCs are differentiated intoosteoblasts and lipoblasts.2. All wound tracks were simple penetrating wounds and were good at stability andrepeat.There was no statistic difference with size of wound track entrance andexit among group A and group B (p>0.05). Their maximum cross-sectional areaswere significantly different (p<0.05). Bacterial count showed that contaminationto the wound track was heavy, and there was no statistic difference among groups(p>0.05).3. With this process, the wound-track bacterial content of group A wassignificantly lower than that of group B (p<0.05). The wound healing time ofgroup IB was also shorter. The pathologic findings of concussion area shows thatinflammatory response in group A were lower than group B. The healing effectsof group A were better than that of group B.Conclusion:1. An effective and ideal isolation and culture system of The hUCMSCs in vitro hasbeen established by tissue explants adherent method. The hUCMSCs wereisolated and purified by this method have high purity and stabile biologicalproperties. It applicates for further research on futher study.2. The hind legs wounded with two kids of projectiles have a similar effect. Butthe group wounded with projectiles fired by a56-type musket (7.62mm) tosimulate real conditions better, more suitable for further study.3. hUC-MSCs is effective in the treatment of firearm wounds. |
