| Objectives:The effects of perfluorocarbon emulsions (PFCE), hypertonic citrate adenine solution(HCA), and both the mixture solution(HCA/PFCE) on oxidative damage, apotosis andpathomorphism and ultrastructure of isolated the kidney of cold storage were investigated. Theaim of study was to search for a safe and effective way to preserve donor kidney.Methods:Male SD rats72, were randomly divided into six groups, and twelve rats in each group, theywere divided into HCA, HCA/PFCE, PFCE. The kidney was infused in situ with thecorresponding HCA solution, HCA/PFCE liquid, PFCE liquid, and rapidly harvested，they werepreserved at o-4℃above20ml of liquids respectively for24and48hours. Samples wereharvested and tested the concentration of MDA, SOD, Bcl-2. The kidney cortex was made toobserve the morphological and ultrastructural changes of renal through light microscopy andtransmission electron microscopy.Results:(1)The concentration of MDA,SOD of kidney cortical cells among preservation solutions ofthree groups has no relation with time (p>0.05);(2) There were significant differences instatistics for MDA, SOD，Bcl-2and pathological score among three solutions (P <0.05)；(3)There were significant differences in statistics of Bcl-2and pathological score between24hoursand48hours (P<0.05);(4) the degree of ultrastructural changes of injury of renal tubularepithelial cell were lighter in HCA/PFCE solution and in PFCE solution than in HCA solutionamong three solutions，and the degree of injury of renal tubular epithelial cell for24hours werelighter than48hours.Conclusion:Perfluororcarbon emulsions on reducing oxidative damage and apoptosis of preservation ofdonor kidney is better than that of HCA/PFCE liquid, so perfluorocarbon emulsion of donorkidney preservation effect is good. |
