ObjectiveTo investigate the potential protective effec of tacalcitol on the human epidermalmelanocytes(MCs) oxidative stress injury induced by hydrogen peroxide, providing atheoretical basis and laboratory basis for the repigmentation treatment of vitiligo.MethodsIn this study, human epidermal melanocytes were cultured in hydrogen peroxide withdifferent concentrations and treatment time, After which cell viability was determined by anMTS assay.Then building melanocyte oxidative stress model in vitro based on the aboveresults.Reactive oxygen species(ROS) levels were evaluated by lasers canning confocalmicroscope. SOD activities and MDA levels assayed by he relevant indicators of oxidativestress testing kitResult1. The human epidermal melanocytes were treated with0.4mM H2O2for24h, the cell viabilitydecreased significantly compared to the control group (P was0.004、0.005).2. Human epidermal melanocytes were pretreated with10-8M、10-9M、10-10M concentrations oftacalcitol for24h, followed by exposure to0.4mM H2O2for24h.The cell viability ofexperimental group1decreased significantly compared to the control group(P<0.05), buthigher than the experimental groups2and3(P <0.05).The rate of melanocytes dendriteschange were higher significantly compared to the control group(P<0.05), but lower than theexperimental groups2and3(P <0.05).3. Experiments groups1,2, and3, which containing blue-green fluorescent cells,weredecreasing with the concentration of tacalcitol elevated, reflecting the level of ROS graduallyreduce with the concentration of tacalcitol elevated.4. SOD activities of experimental group1were significantly higher compared to experimentalgroups3,but MDA levels decreased significantly (p <0.05). Conclusion1. Human epidermal melanocytes viability were decreases as increasing in concentration ofhydrogen peroxide and the treatment time.2.Tacalcitol can reduce the effect of hydrogen peroxide on melanocytes viability and dendrites3. Tacalcitol can decrease the level of ROS induced by hydrogen peroxide in melanocytes.4. Throught increasing the activity of SOD and reducing levels of MDA, Tacalcitol canregulate the oxidative sttress.
