ObjectiveTo study the regulation of tacalcitol on the cell proliferation,adhesion, migration and effect ofthe exppression of mRNA for c-kit on the cultured human epidermal melanocytes, in order toprovide a theoretical basis and laboratory basis for the repigmentation treatment of vitiligo.MethodsWe intervented the cultured human epidermal melanocytes with different concentrations oftacalcitol. The cell proliferation was determined by XTT assay after culture for24h、48h and72h,while the adhesion rate of melanocytes was detected by FN-coated culture plates after culture for72h, the migration of melanocytes were tested by transwell microporous membrane assay afterculture for24h, the expression of mRNA for c-kit of the melanocytes was determined bysemiquantitative reverse transcription PCR after culture for72h.Result1. At the range of10-10~10-6mol/l of tacalcitol showed no significant inhibition of theproliferation on human epidermal melanocytes. when intervented the melanocytes for48h,72hwith tacalcitol that in concentration of10-8mol/l, the proliferation of melanocytes could besignificantly promoted, which has a significant difference compared to the controlgroup(melanocytes without drugs treatment)(P was0.029、0.044).2. At the range of10-8~10-7mol/l of tacalcitol could significantly promote the adhesion rate ofhuman epidermal melanocytes to fibronectin protein (FN), there was a significant difference thatcompared to the control group (melanocytes without drugs treatment)(P was0.003,0.007).3. According to the results of proliferation of melanocytes, we knew each concentration oftacalcitol was without significant toxicity on human epidermal melanocytes, then we selected thecells that co-cultured for24hours for the subsequent migration experiments. The results showedthat at the range of10-9~10-8mol/l of tacalcitol could significantly promote migration ofmelanocytes while compared to the control group (melanocytes without drugs treatment)(P was0.003,0.000). 4. At the range of10-9~10-7mol/l of tacalcitol could significantly promote the expression ofmRNA for c-kit of the melanocytes by semiquantitative reverse transcription PCR when culturedfor72h, there was a significant difference that compared to the control group(melanocyteswithout drugs treatment)(P was0.000).Conclusion1.Tacalcitol could promote the proliferation of human epidermal melanocytes in vitro, enhanceadhesion to fibronectin of melanocyte and promote the migration of melanocytes on fibronectinprotein.2.Tacalcitol could partially raise the expression level of mRNA for c-kit on human epidermalmelanocytes, there was certain correlation in the changes of the expression of mRNA for c-kit tothe cell adhesion and migration. Tacalcitol may promote the ability of cell adhesion, migrationfor the human epidermal melanocyte by increasing the expression of mRNA for c-kit.
