Objective: The main aim of this study was to evaluate the effects of LCP50andLCP50W, polysaccharides extracted from Litchi chinensis, on immnune functions ofKM mice.Methods: At first, we analyzed the effects of LCP50and LCP50W alone oralong with Concanavalin A (ConA) on the proliferation of mouse spleen lymphocytesby MTT method; and then we studied their effects on NK cell cytotoxicity; after that,the ELISA method was used to research their activities on cytokines interferon-γ(IFN-γ) and interleukin4(IL-4) secreted by mouse lymphocytes; next, transcriptionfactors T-bet and GATA-3were further studied by RT-PCR method; further, theiractivities on lymphocytic cycle distribution were measured through flow cytometry(FCM); the effects of LCP50on carbon clearance and delayed type hypersensitivitywere also investigated.Results: LCP50(3.9-250μg/mL) and LCP50W (3.9-125μg/mL) promoted theproliferation of spleen lymphocytes in a concentration-dependent manner; along withConA, LCP50(7.8-250μg/mL) and LCP50W (3.9-125μg/mL) stimulated theproliferation of spleen lymphocytes; they also promoted the cytotoxicity of naturalkiller cell (NK cell) on YAC-1cells at the concentration3.9-250μg/mL in aconcentration-dependent manner; LCP50(7.8-125μg/mL) and LCP50W (3.9-62.5μg/mL) boosted the secretion of IFN-γ, while inhibited that of IL-4; LCP50andLCP50W inhibited the expression of GATA-3while enhanced that of T-bet; From theresults of Flow cytometry, we found that they promoted the development of cell cycletowards s phase. LCP50enhanced the ability of carbon clearance and showed bestactivity at the concentration of200mg/kg; LCP50also promoted the reduced DTHactivity, which showed significant differences at the concentrations of200mg/kg and400mg/kg compared with normal control group.Conclusion: LCP50and LCP50W, polysaccharides extracted from Litchichinensis, have the potential effects of immune enhancement on mice.
