Object: Chronic urticaria is one of the most common skin allergic diseases, itsclinical features including long course, frequent recurrance. This disease seriouslyinfluenced the patient’s work and the quality of life. Recent researches found that thepolymorphism of transformation growth factors have relationship with the occurrenceof Chronic allergic asthmatic disease. However, chronic urticaria which is onemember of Chronic allergic disease family has no correlation transformation growthfactor, the research abroad and domestic was rarely reported. The aim of this study isto examine the characteristic of the genotype distribution of the TGF-β1SNP-509C'T in chronic urticaria patients and healthy controls and to explore the possible rolesof TGF-β1SNP-509C'T in the development of chronic urticaria.Method: The50objects are from Guangzhou first people’s hospital dermatologicaloutpaints, who are clinics typical cases of chronic urticaria. The other50objects inthe control group are on-the-job personnel, who are health. The experimental groupand control group were ruled out transformation growth factors correlated withdisease, no autoimmune disease and family genetic disease. Drawoff2ml peripheralblood, after centrifugal extract DNA, according to the specifications,usingPCR-RFLP and gene sequencing method to detect cases of TGF-β1gene-509C/T site polymorphism the distribution in a chi-square test, in chronic urticaria and normalcontrol group.Result: In object group and the control group, three combination genotype weredetected: CC type, CT type, type TT. TGF β-1-509C/T site genotype frequencydistribution has statistically significant in two groups [ratiorespectivelyTT:CT:CC=8%:28%:64%in CU group,TT:CT:CC=10%:58%:32%ingroup nc, The chi-square=10.892, P=0.04<0.05].TGF β-1-509C/T site allelefrequency has no significant differenc,[CU group C: T=68%:32%, NC group C: T=61%:39%, the chi-square=1.070,P=0.301>0.05], prompting that highexpression of CC gene type may be one of the factors for chronic urticaria.Conclusion:1、 Chronic urticaria polymorphism development will be associated withtransformation growth factor TGF-β1promoter area-509C/T sites.2. Through the experiment, high expression in chronic urticaria group of TGF β1-509-C/C gene locus type prompting that: TGF β-1-509C/T site CC genotypes maybe one of the mutagens for chronic urticaria.
