| The productions of three-line hybrid seeds not only cost low, but was convenient and highlyefficient. It had have many people focus their eyes on it. Recent years, in China, breeding andapplication of three-line hybrid has obtained significant progresses. But there still have some problemson application and superiority combination breeding, such as the influence of external environmentcondition to the fertility restoration ability of restorer, which limited the application and development ofthree-line hybrid cotton to some extent. So it is necessary to explore three-line hybrid cotton. Using3restorers lines of different nuclear background and7cytoplasmic male sterile lines of Gossypiumharknessii and their crosses, which were planted in three different ecological districts, throughcharacters investigation and tested indoor and et al, adopting the statistical analysis technique of ADEmodel and SAS, the study definituded the superiority combination and genetic effect of yield and fiberquality, and explored that the nuclear background of restorer impact on recovery ability and itsrelationship with external conditions such as temperature. In addition, using cytoplasmic male sterilelines of Gossypium harknessii and its maintainer line, restorer and near-isogenic lines(NILs) of restoringgenes,17HSP genes was analyzed by means of real-time fluorescent quantitative PCR. The relationshipof tempreture and fertility was tentative explored. The specific results were as follows:1. Using3restorer lines of different nuclear background and7cytoplasmic male sterile lines ofGossypium harknessii as parents,21crosses were made according to an incomplete dialleldesign(NCâ…¡). These crosses were planted in three different ecological environment. The geneticanalysis showed, yield and fiber traits was significantly effected by additive effects. Yield and yieldtraits were significantly effected by additive interaction effects with environment, and such werefiber length and micronaire which were fiber traits.2. The analysis of restoring capability of ZBRã€H46and H80suggested the results that the fertility ofH46and H80was better than ZBR in both Anyang and Wuwei, and F1whose parent was restorerH46and H80, whether the pollen fertility or yield, had a higher level than whose restorer was ZBR.Restoring capability was related to restorer and the pilots where the materials were planted. F1pollen fertility was significantly correlated with its yields.3. HSP genes expression of differences in the two sets of materials and3restorers was explored bymeans of real-time fluorescent quantitative PCR. The experimental results showed, Ghi_HSP-5ã€Ghi_HSP-8ã€Ghi_HSP-14ã€Ghi_HSP-17have similar expression in the two sets of materials, butexpression of Ghi_HSP-5in restorers was obvious different from the other three and their sourceswere different. Ghi_HSP-1ã€Ghi_HSP-2ã€Ghi_HSP-3and Ghi_HSP-6had a low expression in H46,whose restorer ability was strong. These four genes had the similar expression in other materials.These studies laid a foundation for further study of the mechanism of the restoring genes. |
