| Asparagus,(Scientific name:Asparagus officinalis) is known as Longxucai (refers to the spears of asparagus) in Chinese. Asparagus tastes delicious, and also has high nutritional value and medicinal value. Compared with the organs of diploid plants, polyploid plant organs are much greater in general, due to the edible part of asparagus is its spears, which is an important factor in the increase of production to improve asparagus, the asparagus polyploid breeding is of great significance.This experiment used the whitish seeds and seedlings of "Jinggang701" diploid asparagus which is mainly planted in the South of China as experimental material. The polyploidy asparagus was induced with colchicine, and the successfully induced tetraploid asparagus was conducted with tissue culture. The experiment has achieved the following research results:1. Study the influence to the induction effect of colchicine concentration and centrifugal rate when immersing. The results showed that:using0.2%colchicine to immerse the whitish seeds for6h would work best. The highest tetraploid induction rate was20%, but the influence of different centrifugal rates when immersing was not obvious; while using0.3%colchicine to drip the seedlings continuously for4d, the highest tetraploid induction rate was18%. The result indicated that the induction effect of immersing whitish seedlings was better than dripping the seedlings.2. The experiment compared partial resistance and morphological characteristics of the tetraploid asparagus and the diploid asparagus. The results showed that the leaves longer and thicker, stems thicker than the diploid.3. The comparison of partial resistance between tetraploid asparagus and diploid asparagus showed that the tetraploid were significantly better than the diploid in terms of heat resistance and drought resistance.4. By identification of flow cytometry and cell chromosome:the chromosomes of tetraploid asparagus from the20becomes40; Redouble plants also appeared the chimeras of diploid and tetraploid as well as the chimeras of diploidã€tetraploid and the octoploid.5. The experiment induced and planted the regeneration plants of the tetraploid asparagus and the results showed that:the optimal culture medium for tetraploid asparagus stem tip to induce callus induction was:MS+2.5mg/L2,4-D+1.5mg/L KT+25g/L sucrose+6.5g/L carrageenan (pH5.8); the optimal culture medium for callus induction to proliferate, induce and differentiate adventitious buds was:MS+ 1mg/L NAA+1.5mg/L6-BA+30g/L sucrose+7g/L carrageenan(pH5.8)ï¼›the optimum culture medium for rooting was:1/2MS+3mg/LPP333+0.5mg/LIBA+30g/L sucrose+7g/L carrageenan(pH5.8)... |
