White spot syndrome virus (WSSV), a large, non-occluded, enveloped, rod-or elliptical-shaped, doubled-stranded DNA virus, belongs to the Whispovirus genera, Nimaviridae family. It is one of the major viral pathogen of cultured shrimps due to its strong pathogenic ability and wide transmission range. However, there is still no effective methods to control or prevent it. Proteins encoded by immediate-early (IE) genes play vital roles in the virus life cycle. wsv083is an immediate-early gene of WSSV that predicted as a Ser/Thr protein kinase. The target of viral protein kinases may be virus or hosts proteins. VP28and VP19, the main membrane protein, undergo threonine phosphorylation modification. Does the WSV083modulate VP28and VP19phosphorylation? So, we carried out our research and found some results as follows. First of all, we proved WSV083can not modulate VP28and VP19phosphorylation in vitro, second, we proved WSV083can auto-phosphate its serine, threonine and tyrosine residues, and it is a DSK (Dual-specificity protein kinase). Third, The S134, T66, Y165and Y304of WSV083are the sites of serine, threonine phosphorylation respectively. At last, WSV083is located in nucleus and as point-like. However, the mutant of WSV083which lose kinase activity also is located in nucleus without point-like.We predict that the target of WSV083is involved with its catalytic activity.
