As a fine mid-bodied species,Wanxi White goose has the characters of seasonal egglaying and high broodiness,it strongly inhibit its egg production.So it probably be the mosteffective way to improve reproduction of Wanxi White goose throuht eliminatebloodness.PRL plays a great role in bloodness occuring and keeping in avian species.Forthis purpose, Wanxi-White goose was choosed to be researched object in thisexperiment.RPRL is prepared in laboratory and Wanxi-White goose was activelyimmunizatied through neck subcutaneous injection.Finally,the results were analyzed thatthe level of reproductive hormones effects on egg production in the body.The resultsshowed that:1.PRL genes of Wanxi-White goose were constructed into pET32a-PRL andPet32a-PRL was expressed though induction of IPTG.Then recombinant proteins hadmolecular weight approximately to45kD and262.5μg/ml of expression amount.In thisway,RPRL protein accounted for about35.2%of the total protein.Recovery of His-tagcolumn was about64.9%.2. Through twice immunization PRL,FSH,VIP level increased. GnRH,-EP leveloverall reduce firstly and then increase while LH decreased.PRL level: Through twice immunization,groups of PRL levels were elevated, andPRL levels had no significant difference between five groups. FSH levels: Through firstimmunization, group I (23.42±2.80Ang/ml) was significantly elevated compared with thenegative control, positive control (p<0.01), group II (19.81±10.8ABng/ml) increased morequickly than the negative control, positive control groups (p<0.01); Through secondimmunization, the level of FSH in serum rised in each group, with no significant differencebetween the groups content. GnRH levels: Through first immunization, group I rised, andthe other four groups decreased in different degrees. Through second immunization, allgroup contents increased with no significant difference.LH levels: Through firstimmunization, level of each group reduced.Then through second immunization group IIcontent rised while the other four groups reduced with no significant difference betweenfive groups.VIP level:Through first immunization groupI(128.3±73.8Apg/ml) had verysignificantly higher level(p<0.01)compaed with positive control.Through secondimmunization, level of four groups rised with level of groupⅢ decreased withoutsignificant difference.-EP level: Through first immunization, level of group I (355.3±241Ang/L) rised significantly (p<0.01)with the other four groups decreased withoutsignificant difference. Through second immunization, level of the each group rised without significant difference.3. Active immunization of recombinant PRL proteins can reduce the rate of WanxiWhite goose bloodiness.Egg production had no significant difference.
