In this paper,the methods of strains preparation for Cordyceps militaris was studied,including tissue separation and spore separation.By observing and determining the mycelial growth status,color conversion performance and fruit body growth and yield to explore the best way of the preparation of C.militaris strains.The different kinds of carbon sources(glucose,sucrose,maltose,lactose)and nitrogen sources(ammonium sulfate,peptone,yeast powder,urea)are selected and designed in the concentrations of different carbon and nitrogen source to make PD liquid medium to cultivate C.militaris liquid bacteria,and the C.militaris fruit body is cultivated.By determing the biomass of liquid strain,the fresh weight of fruit body,the polysaccharide and cordycepic acid content of C.militaris and spent cordyceps substrate to research the effects of different carbon and nitrogen sources on the growth and quality of Cordyceps militaris.The results show:1.During the growth of 10 d and 15 d C.militaris for tissue separation,the mycelium of C.militaris grows fast and the color changes early after illumination;Selecting the growth of 15 d C.militaris for tissue separation,the liquid bacteria is fine and viscous,which is more ideal.Cultivation of 40 d C.militaris for tissue separation,the yield of fruit body is significantly higher than other periods(P<0.05),and the average yield of C.militaris is 22.34 g.2.During the growth of 45 d C.militaris for spore separation,the cultured liquid mycelium ball is the most ideal,the cultivation of the resulting sub-solid yield significantly higher than other periods(P<0.05),each box reached 20.15 g.So the growth of 45 d C.military is more suitable for spore separation.3.Adding different kinds and concentrations of carbon sources and nitrogen sources had a significant effect on the dry weight of C militaris liquid bacteria cultured in PD(Potato Dextrose)liquid medium(P<0.05).When the carbon source is 30 g/L sucrose and nitrogen source is 6 g/L yeast powder,the mycelium dry weight is the largest,1.01 g/100ml and 1.11 g/100ml respectively.Adding different kinds and concentrations of carbon sources and nitrogen sources in PD liquid medium to culture of liquid bacteria to cultivate C.militaris,the yield of C.militaris have significant differences(P<0.05).Adding 20 g/L sucrose as carbon source and 10 g/L ammonium sulfate as nitrogen source cultured liquid bacteria,the yield of C.militaris is the highest,and average fresh weight is 24.46 g and 25.6 g,respectively.4.Different kinds and concentrations of carbon sources and nitrogen sources liquid bacteria cultured from nitrogen sources had a significant effect on the content of cordyceps polysaccharide and cordyceps acid in C.militaris and spent cordyceps substrate.10 g/L glucose,12 g/L peptone as the source of carbon and nitrogen of liquid bacteria respectively,cultivated C militaris and its cordyceps polysaccharide content is high,1.81 g/100g and 1.91 g/100g respectively;The higher polysaccharide content in the spent cordyceps substrate was 30g/L sucrose and 12g/L ammonium sulfate,0.07 g/100g and 0.04 g/100g respectively.Using 25g/L maltose and 6g/L Peptone as the source of carbon and nitrogen of liquid bacteria respectively,the content of cordycepic acid in C.militaris was higher,4.71%and 5.48%,respectively;The higher content of cordyceps acid in spent cordyceps substrate was 30 g/L glucose and 8 g/L peptone,which were 2.09%and 2.29%,respectively. |
