Polyploid Induction And Identification Of Dendrobium Officinale

A preliminary study on the polyploid induction of Dendrobium officinale in this paper, thematerials include seeds, embryo, PBLS and stems of Dendrobium officinale, the inductive agent iscolchicine, the ways are soaking in different concentrations of colchicines and adding colchicines inmedium. Trough the methods to identify polyploidy plants, such as Flow cytometry, counting thenumber of chromosome. etc. A batch of polyploidy materials were sifted, this laid the foundation forfurther study on polyploidy Dendrobium officinale. Main research results are as follows:1、The materials were treated by soaking method, which include embryos, PBLS and stems ofDendrobium officinale. With the rise of the concentrations of colchicines, the death rate of the materialsis rising. From the point of mutagenic effects, all the processes can get polyploidy. But the inductiveeffect of embryo is the best, when the concentration of colchicines is0.3%, the time is36hours, theinductivity is the highest as40%, and more tetraploid plants were discovered.2、The materials were treated by adding colchicines in the medium, which include seeds andPBLS of Dendrobium officinale. Taking seeds as material, when the concentration of colchicines is50mg/L, the inductivity is50%, and most of them are tetraploid.3、The polyploid plants of Dendrobium officinale have the following characteristics, such asdwarf, cluster, leaves have clearly stripes, thickening, bending, bottle green, fragile, easy broken and soon.4、 By counting the number of chromosome, the diploid Dendrobium officinale has38chromosomes, while the tetraploid has76chromosomes.In view of the polyploid induction of Dendrobium officinale, this research found that taking seedsof Dendrobium officinale as material, through the method of adding colchicines in medium, when theconcentration of colchicines is50mg/L, the time is4months, the mutagenic effects is the best. So thisis the best way to induced polyploid of Dendrobium officinale.
