Haynaldia villosa (syn. Dasypyrum villosum L.,2n=14, VV) is annual cross-pollinated herb, a wild relative of common wheat and natives to the Mediterranean coast and the Caucasus. It resistants to powdery mildew, rusts, eyespot, wheat streak mosaic virus and other diseases, as well as more spikelets, frost and drought tolerance, high grain protein content features. Wheat-H, villosa translocation with small alien fragment is considered as an important genetic resource for wheat improvement. To locate the powdery mildew resistance gene Pm21, create more involved6VS different sections of the structural variants and induce powdery mildew resistance translocation lines with more small chromosome segment of H.villosa. The mature female gametes of wheat-H.villosa6VS/6AL translocation line92R137was selected as radioactive materials. Molecular cytology approach was used to detect the chromosomes structural changes involving6VS of H. villosa. Common wheat lines carrying single segment of H.villosa chromosome6VS were screened by genomic in situ hybridization. At mitosis metaphase of root-tip cell of irradiated materials was used to detect the chromosomes structural changes involving6VS of H.villosa by GISH.51alien lines containing chromosome structural changes with small segment of6VS were identified, including19interstitial translocation lines,20terminal translocation lines,8deletion lines and4multiple translocation lines.By comparative genetic mapping analysis, there is a linear relationship between the chromosomes of wheat homoeologous group6and rice chromosome2.95pairs of primers were designed by comparative genomics. Chinese spring, H.villosa,92R137, Del.6VS-1and Del.6VS-2were choosen to screen and locate these markers by PCR.20specific molecular markers for6VS were founded, including8markers reported by others in our laboratory. Through molecular marker analysis,31structural variants have been recognized for their fragment size and breakpoind. They are very useful for constructing structural aberrance library and physical maping. The Pm21gene was further located in the region between FL:0.45and FL:0.58of6VS.In the present study,27homozygous lines were selected.13of them could judge its wheat background by FISH using repetitive DNA sequences676D4, pSc119.2and pAsl as probes.
