In this study, PLIN1and RBP4genes which are candidate genes about animallipid metabolism regulation were selected, its SNPs (single nucleotide polymorphisms)were detected and associated with carcass and meat quality traits were calculated infour Anhui superior native pig breeds(Anqing Liubai Pig, Huoshou Black Pig,Wannan Black Pig and Wei Pig). Primers were designed according to Sus scrofa’PLIN1(Number:NC010449)and RBP4)Number:NC010456)gene whichpublished in NCBI, polymerase chain reaction-single-strand conformationpolymor-phism (PCR-SSCP) and sequencing were used to detected the SNPs andanalysis of GLM model in SPSS program were used to detected the genetic variationand its associated with carcass and meat quality traits were calculated. In order to findthe effective genetic markers of carcass and meat quality traits in native pig, andprovide reference of molecular biology for the study of germplasm characteristics andMAS in Anhui native pig breeds.The study show that:(1) There was no mutation in exon3,4,5,6,7and intron4in PLIN1gene inthese four populations;Moreover there was no mutation in exon3,4,5,6and intron3in RBP4gene in these four populations.(2) The results showed that An A→G mutation in exon2at3519bp, the amino acidsencoded by histidine to arginine.3genotypes (AA, AB and BB) were found in PLIN1gene in Anqing Liubai Pig and Wannan Black Pig. The genotypic frequency of AAwas29.09%,8.47%respectively, AB was57.27%,43.22%respectively and BB was13.64%,48.31%respectively, the polymorphism information content (PIC) was0.3690in Anqing Liubai population and0.3322in Wannan Black population, theywere all in average polymorphism situation. But there was no mutation in exon2inHuoshou Black Pig and Wei Pig populations, it is wild genetype. The results showedthat An A→G mutation in intron3at604bp and3genotypes (CC, CD and DD) werefound in RBP4gene in Anqing Liubai Pig, Wannan Black Pig and Wei Pig. Thegenotypic frequency of CC was53.64%,66.10%,57.28%respectively, CD was37.27%,28.81%,31.07%respectively and DD was9.09%,5.09%,11.65%respectively, the polymorphism information content (PIC) was0.3205in AnqingLiubai population,0.2646in Wannan Black population and0.3175in Wei Pigpopulation, they were all in average polymorphism situation. But there was nomutation in intron3in Huoshou Black Pig populations, it is wild genetype.(3)χ2test showed that the gene frequencies and genotype frequencies havereached the Hardy-Weinberg equilibrium (P>0.05) in Anqing Liubai Pig and WannanBlack Pig in PLIN1gene; the gene frequencies and genotype frequencies havereached the Hardy-Weinberg equilibrium (P>0.05) in Anqing Liubai Pig, Wannan Black Pig and Wei Pig in RBP4gene.(4) The associated of polymorphism of PLIN1gene with carcass and meatquality traits were analysed. In Anqing Liubai Pig, AA and AB type was higher thanBB type in average backfat thickness (P<0.01); AA and AB type was higher than BBtype in leam percentage (P<0.05); BB type was higher than AB type in watercontaining (P<0.05); AB type was higher than BB type in the intramuscular fat(P<0.05).In Wannan Black Pig, AA type was higher than AB and BB type in averagebackfat thickness (P<0.01); AA type was higher than BB type in loin muscle area(P<0.05); BB type was higher than AA type in the intramuscular fat (P<0.01), higherthan AA type(P<0.05); AA type was higher than AB and BB type in tenderness(P<0.01).(5) The associated of polymorphism of RBP4gene with carcass and meat qualitytraits were analysed. In Anqing Liubai Pig, CC type was higher than DD type inaverage backfat thickness (P<0.05); CC type and CD type was higher than DD type inloin muscle area (P<0.05); CC type was higher than DD type in a*value (P<0.05);CD type was higher than DD type in average backfat thickness (P<0.05). In WannanBlack Pig, CC type was higher than DD type in average backfat thickness (P<0.05);CD type was higher than DD type in b*value (P<0.05).In Wei Pig, CC type washigher than CD type in a*value (P<0.05).The study suggest that PLIN1gene was used as candidate genes in MAS whichinfluence carcass and meat quality traits in Anqing Liubai Pig and Wannan BlackPig;And RBP4gene as candidate genes in MAS which influence carcass and meatquality traits in Anqing Liubai Pig, Wannan Black Pig and Wei Pig.
