Detection of pathogens is the basis for prevention and control of diseases in aquaculture.Aptamers have a great potential application in the diction of microorganisms for their numerousadvantages like high affinity, strong specificity. Based on the aptamers selected previously andthe high sensitivity of PCR, the present thesis established a qualitative detection technologytargeting the aquaculture pathogens Vibrio harveyi and Vibrio alginolyticus.Based on the qualitative detection technology, three aptamers(S12、S25、S36) against V.harveyi and five aptamers (1#、11#、35#、37#、41#) against V. alginolyticus were studied and usedto detect the two pathogens in the present thesis. The results showed that the aptamers S12hadbetter specificity and sensitivity, and could detect1.55×101cells/ml V. harveyi. Aptamer S36also had good specificity and sensitivity, and could detect1.55×102cells/ml V. harveyi, butaptamer S25failed in the present qualitative detection. In the detection of V. alginolyticus,aptamers37#,1#and11#showed better specificities and sensitivities, and could detect1.16×101cells/ml V. alginolyticus; aptamer41#could detect1.16×102cells/ml V. alginolyticus; butaptamer35#failed to detect V. alginolyticus in the present experiment.
