Fodder cost is the biggest expenditure item in broilers breeding production costs,accounting for more than70percent of the total cost of the whole chicken farming. Aserious impact of the economic efficiency of broilers of the Broiler feeding andmanagement is not strictly in the actual production process, the fodder with the unscientificuse, resulting in a waste of fodder, especially in the case of the protein raw materialshortages and the rising prices.Microbial agents as a new type of feed additives can to promote animal growth,improve feed conversion rate and enhanced the role of the the animal immune organfunction. Bacillus as a class of viable bacteria microbial agents not only has the balance ofthe regulation and control gastrointestinal flora, to improve animal body’s own immunesystem to improve the role of animal production performance, but also has a strong stressresistance,high temperature resistance, high voltage resistance and other uniquebiologicalcharacteristics, has a strong producing protease, amylase, cellulase activity, capable ofdegrading to the plant as a source of macromolecules, such as cellulose, starch, andproteins, etc., as well as the feed of the certain anti-nutritional factors,in order to promoteanimal digestion and absorption of the nutrients, so as to achieve the purpose of improvingthe feed conversion ratio.This trial were obtained from28-day-old healthy chicken jejunal mucosal sporeability and with strong production protease strain P-98, producing of cellulase strain C-8,producing amylase strain S-4,and three strains of the colonies, thallus, and sporemorphology observation, physiological and biochemical identification and construction of16S rDNA phylogenetic trees, the strain P-98was initially identified as Bacillusmethylotrophicus; the strain C-8was initially identified as Bacillus methylotrophicus; thestrain S-4was initially identified as Bacillus velezensis.In order to the enzyme production agents for the late broilers feeding trial. In vitroevaluation of biological characteristics of the enzyme producing Bacillus, The results showthe screening of the three bacteria have a faster growth performance and enzyme producingability, the ability to inhibit the the harmful bacteria; has excellent of low pH values, bile salts and high temperature tolerated; And through the mice and chicks gavage test reflectsit has a good security.In order to carry out the production of the three strains inoculants we were sporeforming condition optimization, Through the single factor experiment and orthogonalexperiment the results shown that,The spore forming fermentation optimum conditions forthe strain S-4is: corn meal in2.0%, corn steep in1.5%, NaCl in0.2%K2HPO4in0.4%,KH2PO4in0.1%, the initial pH value in7.0,250mL triangular flask liquid volume50mL,inoculum size in6%, shaking speed in180rpm, the incubation time is48hours,fermentation temperature in37°C; The spore forming fermentation optimum conditions forthe strain P-98is: starch in0.5%, yeast powder in0.5%, MnSO4·H2O in0.1%, K2HPO4in0.4%, KH2PO4in0.1%, the initial pH value in7.0,250mL triangular flask liquid volume50mL, inoculum size in6%, shaking speed in180rpm, the incubation time is48hours,fermentation temperature in37°C; The spore forming fermentation optimum conditions forthe strain C-8is: corn meal in2.0%, soybean peptone in2.0%, MgSO4·7H2O in0.05%,CaCl2in0.02%, Na2HPO4in0.2%NaH2PO4in0.1%, the initial pH value in7.0,250mLtriangular flask liquid volume50mL, inoculum size in6%, shaking speed in180rpm, theincubation time is48hours, fermentation temperature in37°C. Strain P-98fermentationbroth spores yield reached87.60%; strain C-8fermentation broth spores yield reached95.20%; strain S-4fermentation broth spores yield reached95.60%. Laid the foundationfor the production and application of strains of the spore microbial agent.Through the results of the optimization of fermentation conditions to production thethree strains agents, for a42-day broilers feeding experiment. And counted the numbers ofE. coli, Lactobacillus, Bifidobacterium in fecal samples by GB. Determination the changesof protease, cellulase, amylase, lipase vigor in the duodenum, And calculation of thepercentage content of organic constituents and phosphorus and calcium in the fecalsamples.In the process of the feeding, Each experimental group duodenum digestive enzymeactivities have been significantly improved as compared with the control group. And allkinds of nutrients residual in the feces has reduce. Digestive enzymes produced by Bacillusindeed contribute to of the decomposition of nutrient, and promoted the absorption.With increase of the usage time of Bacillus, the intestinal microflora of each dealingcompared with the control group, there was a downward trend about E. coli, Lactobacillusand Bifidobacterium was a elevated trend, that Bacillus can regulating intestinal microecological balance. Enhanced the immune organ index and effectively suppress of thechick intestinal’s E. coli and other harmful bacteria growth and reproduction. Improve theconcentration of the lactobacillus beneficial bacteria, and improve the tract environment ofchicks.Consolidated the results of the broilers feeding trials show that, it can rapidly improve the intestinal enzyme activity after feeding enzyme producing of Bacillus, the broilersfeeding experiment of three strains has reached a good effect, this three kinds sinensis hasa role in the prevention and treatment of broiler diarrhea, enhance immune organ index,improve feed conversion rates, improve growth performance. And to provide thetheoretical basis of the scientific by improve feed efficiency, reduce production costs,improve economic efficiency.
