The variations of the Adiponectin gene was detected and analysed in chickensfrom the F2resource population by PCR amplification,DNA sequencing,PCR-RFLPand CRS-PCR-RFLP,and the assoiation analysis of the polyphorisms in the geneswith the economic traits was performed using the least-squares analysis. The mainresults were as follows:1.A total3polymorphic loci were pick out for the first time in the study of thisgene from a DNA pool was build using DNA samples from72chickens of F2resource population of Gushi chicken crossed with Anka broiler.2. Three SNP locus G318C, T684C and A976G were first detected by theapplication of PCR-RFLP technique based on the5’UTR sequencing results.Sequencing results showed that three SNP loci were G/C mutation, T/C and G/Amutation respectively. Former two loci have two alleles and three kinds of genotype.A976G had two alleles, two genotypes. Genetic variation analysis found that G wasthe dominant allele (0.73) and GG genotype (0.45) is the predominant genotype inG318C locus. In T684C locus, C was the dominant allele (0.72) and the predominantgenotype was CC genotype (.54). In A976G locus, A was the dominant allele (0.627),and the predominant genotype was AG genotype (0.746).3The detected three SNP loci were conducted association analysis witheconomic traits. The results showed that G318C mutation of adiponectin gene hadsignificant correlation with total protein, globulin,4weeks old pelvic width,12weekschest width and8weeks sternum length; T684C mutation had significantly associatedwith head weight ratio and γ-glutamyl transferase peptide enzymes; A976G mutationhad significantly correlated with subcutaneous fat thickness, LDL, pancreatic weight,breast muscle fiber roundness of total cholesterol and high density lipoprotein. Therefore, it can be reasonably concluded as follows:The5’UTR region of chicken APM1is rich in genetic variation, implying a greatroal in various biological processes such as post transcriptional regulatory pathways,stability and translational efficiency.There polymorphism sites were significantlyrelated with some economic traits of chicken,they may be worked through the changeof mRNA stability and decomposition rate and give further influence on the geneexpression. These polymorphism sites can be expected to develop possible molecularmarkers for improve the flavor of chicken meat and sellection of corresponding traitsin the chicken breeding. Further investigations are required for detecting theunderlying machinim of these polymorphism site on the gene function.
