| This paper was to study the genetic diversity’s analysis between wild lentinus edodes strains from Hot-Dry valley area and daily production strains mainly.To research whether it have great value in breeding; genetic diversity’s analysis was mainly through four parts to study the mushroom.First of all, amplifying sequence of swab strains by ITS PCR, electrophoresis and geting electrophoresis image.according to the MAKER to determine whether the tested strains were lentinus edodes.then sequencing and construction of phylogenetic tree.we can see that only a few wild strains have a high similarity coefficient with the production strains;but most of the wild strains have a lower similarity with producting strains.it has a very higher value,also could be used as a treasure house of breeding materials,and would lay a great foundation in breeding.Isozyme analysis was based on mobility to insure the postion of the bands,then use the relative sofeware to got the clustering figure.Geting8strains of lentinus edodes in the cluster figure,such as LDT2012-8,LDT2011-12,LDT2012-6,4MMO5,LDT2011-5, LSM6,LDT10-1,ZMXD04,they have much closer genetic relationship between them and the producting strains.the similarity between those three strains asLDT10-1,4MMO5, LDT2011-5and the producting strains of up to more than80%.it suggest that these strains had great respection,and lay a firm theoretical foundation in the future’s breeding.RAPD analysis was to amplificating sample by10randon primers, electrophoresis and get71bands in total.2of them are the commom bands, polymorphic bands with69. polymorphic bands according for97.2%of the total number of amplified bands.Then through the NTYSY software to analysis band dates,and got the cluster figure.From the cluster figure we can got the result that5MMO2,LVM20101,LDT2012-4,L-LDMT,LDT12-3,LDT2012-8,LDT2011-9,LDT2012-6,LSM-1,L DT2011.16,LDT2012-8,4MMO5,LDT10-1,LDC21020,LDT2011-9those17wild strains have a great value in breeding.besides, LDT10-1,LVM20101,LDT2012-4,L-LDMT, those4strains have a higher similarity between them and the production strains in the same group more than80%.so those strains cab be the breakthrough in the next breeding job. ISSR analysis was to amplificate sample DNA by random primers in vtro, Random primers binding with diffirent sites produce many polymorphic DNA fragments.Using the NTYSY software to process the strip date,and got the cluster map.it can be found by analysising thecluster map that LDT20120-1-2,LDT2011-9,LDT14, L-LDMT,LDT8-2,LDT10-1,4MM05,LDT9-1,2MXD04,135,5MMO2,LDTorLSM2,LDT2011-16,LS M-1,LDC21020,1032. Those strains had a much greater value.In a word, LDT2011-9, L-LDMT,LDT8-2and LDT10-1those4strains will be a great value in the future’s breeding job. |
