Objective:Study on plasma pharmacokinetic of ivermectin following oral administration under different conditionsin Cherry Valley Ducks and in order to validate the effect of pure traditional Chinese medicine "Bao gan hu shen tuo mei su" on preparing aflatoxin B1poisoning,use syndrome differential pharmacokinetics method to study the pharmacokinetics of ivermectin in Cherry Valley ducklins.Method:Four groups of one hundred and fifty-six Cherry Valley Ducklins each were located.The group I (normal group) were in optimal nutritional condition for28days.The group Ⅱ(model group) were in optimal nutritional condition seven days,then the Cherry Valley Ducklins were fed aflatoxin B1contaminated diets for the next fourteen days,and the next seven days the Cherry Valley Ducklins were in optimal nutritional condition.The group Ⅲ(montmorillonite group) were in optimal nutritional condition seven days,then the Cherry Valley Ducklins were fed aflatoxin B1and montmorillonite contaminated diets for the next fourteen days, and the next seven days the Cherry Valley Ducklins were in optimal nutritional condition. The group Ⅳ (traditional Chinese medicine antisyndrome group) were in optimal nutritional condition seven days,then the Cherry Valley Ducklins were fed aflatoxin B1contaminated diets for the next fourteen days,and the next seven days the Cherry Valley Ducklins were in optimal nutritional condition,meanwhile each ducklins oral administrated the solution of preparing "Bao gan hu shen tuo mei su"5ml for7days.Ducklins in each group oral administrated the solution of ivermectin formulated in propylene glycol.On29day.Blood were collected before treatment and up to timing after treatment.The plasma samples were analyzed by high performance liquid chromatography (HPLC) using fluorescence detection.The resulting data were analyzed by DAS2.0pharmacokinetic software.Result:The assay was validated from0.5to100ng/ml.Good linearity was observed in the entire concentration range.The limit of quantification was0.5ng/ml.Regression ofoverall mean recovery value was92.67%.For the normal group, the montmorillonite group and traditional Chinese medicine antisyndrome group,the drug concentration-time data were fitted to a one-compartment model with first order absorption after a single oral administration of ivermectin.For model group the drug concentration-time data were fitted to a two-compartment model with first order absorption after a single oral administration of ivermectin.The main pharmacokinetic parameters of each group was as followrNormal group:Tmax was2±0h, Cmax was49.167±2.439ug/l,AUC(0-∞) was308.534±13.779μg/l·h, Vd/F was3.923±1.017L/Kg,Cl/F was0.649±0.029L/h/Kg,T1/2ka was0.591±0.265h, T1/2ke was4.215±1.191h.Model group:Tmax was5.333±1.155h, Cmax was60.357±13.794μg/l, AUC(0-∞) was710.966±144.139μg/l·h,Vd/F was1.861±0.578L/Kg,Cl/F was0.29±0.066L/h/Kg,Tl/2ka was1.923±1.117h, T1/2β was15.64±15.287h. montmorillonite group: Tmax was6±0h,Cmax was39.137±2.176μg/l,AUC(0-∞) was355.255±34.671μg/l·h,Vd/F was4.604±1.563L/Kg,Cl/F was0.567±0.057L/h/Kg,Tl/2ka was1.708±1.334h, Tl/2ke was5.787±2.441h. traditional Chinese medicine Anti-syndrome group: Tmax was4±2h,Cmax was38.867±11.582μg/l,AUC(0-∞) was323.82±56.454ug/l·h,Vd/F was5.721±0.443L/Kg, Cl/F was0.63±0.106L/h/Kg,Tl/2ka was1.119±0.217h, T1/2ke was6.455±1.439h.Conclusion:This method can be successfully used for the detection of ivermectin in plasma.Ivermectin following oral administration were widely distributed in the Cherry Valley ducks.Aflationg B1can cause hepatic injury and change the pharmacokinetic of ivermectin following oral administrated in Cherry Valley ducklins.It showed slow absorption,weaken absorption degree and weaken elimination rate.But after montmorillonite and antisyndrome by pure traditional Chinese medicine,the rate of absorption and elimination were increased,and there was no significant difference.From the view of pharmacokinetics was to validate the effect of pure traditional Chinese medicine(preparing the hepatic and kidney by toxin off) on repairing the damage which caused by aflatoxin B1.
