The liposome agents as the fourth generation of medicine product has unparalleled advantages. To bring into full play ivermectin efficacy of medicine, or to extend the effective blood concentration time reducing drug consumption, increasing drug treatment index, lower drug toxicity, and changes in administration to avoid the emergence of purposes.The preparation phase of this study is the high encapsulation efficiency, the stability of good treated liposomal ivermectin agents, research in the toxicology, pharmacokinetics and clinical treatment of liposomal ivermectin in the animal body.To optimize the preparing technique and prescription, ivermectin liposomes were prepared by the thin-film hydration and ultrasonic technique to investigate the effect of velocity ,organic solvent and vibrated speed of aether on its encapsulation efficiency and stability ,the optimized preparing techniques were determined:aether as organic solvent, velocity 150 r/min and vibrated speed of eather ,50-100 r/min. The optimum prescription for preparation of ivermectin liposome were selected with orthogonal design: Lecithin:cholesterol=1:0.1; lvermectin: (Lecithin+cholesterol)=1:10 ; PH of PBS is 7.0 , organic solvent: PBS =4:3.The results show: the maximum encapsulation efficiency of prepared ivermectin liposomes has attained (83 ± 1.6) % on the whole steady.In order to study acute toxicity of liposomal ivermectin , Ivermectin liposomes were supplied to each group by i. p in mice , and compared with the ivermectin injection. The acute toxicity tests indicate, the LD50 (92.1516 mg/kg.bw) of liposomal ivermectin set show high the LD50 (24.2493 mg/kg.bw) of invermection injection , is the ivermection injection of 3.8.100 KM mice were randomly divided into 5 groups .2,6,3,0 mg/kg.bw Liposomal invermection and 3 mg/kg.bw ivermection injection were supplied ivermectin injection were supplied to each group by i.p for 4 weeks in succession in order to investigate its subchronic toxicity.
