To evaluate the quality traits selecting of Common carp (Cyprinus carpio L.) and theclarification of HUFA biosynthetic mechanism, we analyze the Correlation betweenmicrosatellite markers with fat content and protein content of muscle. The genotypes of thesecorrelative loci captured were determined.To study the regulatory role ofΔ6FAD and ELOVL5in the biosynthesis of HUFA, the full-length ELOVL5cDNA of mirror carp was cloned by usingreverse transcript PCR (RT-PCR) and rapid amplification of cDNA ends (RACE).The enzymegene expression in different tissues of Mirror carp was determined. It will be helpful for clarifythe HUFA biosynthetic mechanism in Common carp, for the development of method inenhancing such ability. The latter will accelerate the development and application of formulatedfeeds, as well as the development of aquiculture. The results were shown as follows:A total of233microsatellite markers were used to analyze the genotypes in the referencepanel contained107individuals obtained from mirror common carp (Cyprinus carpio L.) strain,and two muscle quality traits including fat content and protein content were tested using GLM(General Linear Model) single marker of the regression. The determination of the thresholdvalues by10000Permutation test revealed that17markers had significant correlation (P <0.05)with the two traits, in which HLJ030, HLJ2560, HLJ3633and HLJ3554had very significantcorrelation with muscle protein content (P <0.01). In addition, the genotypes of these correlativeloci captured were determined by Daucan’s in SPSS17.0software. These functional markers andgenotypes related to the traits may provide the efficient evidence for improving muscle qualitytrait in the common carp.The cDNA for Δ6fatty acid desaturase (Δ6FAD) gene was1471bp in full length andencodes a peptide with444amino acids (accession no. KJ576791). The protein sequence hadcharacteristics of microsomal fatty acid elongases and shared65%-91%sequence identity withother fish’s Δ6or Δ5desaturases gene.The cDNA for fatty acid elongase5(ELOVL5) gene was1121bp in full length and encodesa peptide with291amino acids (accession no. KF924199). The protein sequence hadcharacteristics of microsomal fatty acid elongases and shared74%-93%sequence identity withother fish’s ELOVL5gene.The enzyme gene expression in different tissues of Mirror carp was determined usingreal-time quantity PCR (RT-qPCR). Δ6FAD gene expresses the highest in liver, followed bydorsal muscles、abdominal muscle and brain. Whereas their expression levels in spleen, heart andgill was low or undetectable. ELOVL5gene expresses the highest in liver, followed by brain,and the lowest in muscle.
