The Genetic Diversity Of Several Common Carp (Cyprinus Carpio L.) And Screening Molecular Marker Assisting Breeding For Songpu Mirror Carp

In this article, we made correlation analysis on the body weight, body size and tail size of the 194 Songpu mirror carp by utilizing SPSS 13.0 software. And then we screened 30 microsatellite markers which had been identified well to determine the effective number of alleles (Ne), observed heterozygosity (Ho), expected heterozygosity (He) and polymorphism information contents (PIC) of the new population of Songpu mirror carp as well as checked the population Hardy~Weinberg equilibrium by chi~square test. Eventually, we also analyze the relations of these 30 microsatellites in different genotypes on body weight, body size and tail size. The results showed that there was extremely significant correlation between body weight and body size, and significant correlation between tail size and body weight, body size. The genetic diversity indices revealed that a total of 172 alleles were found, the allele size ranging from 115 bp to 432 bp. the effective number of alleles (Ne) was 1.14 to 5.32, the observed heterozygosity (Ho), the expected heterozygosity (He) ranged from 0.07 to 0.92 and 0.13 to 0.91, and the PIC was 0.12 to 0.79, respectively. Among them, the high (PIC≥0.5) and moderate (0.25≤PIC≤0.5) polymorphic markers were 21 and 7, respectively, which indicated that the genetic diversity and polymorphic information content of this population were high and rich. Meanwhile, we still found that four genetic locus HLJ328, HLJ693, MFW29 and MFW48 had a significant correlation with body size (P<0.05), At the same time, the HLJ328, HLJ693 and MFW48 were also significantly correlated with body weight character (P<0.05), however, There were no loci correlated with tail size. In the study, the microsatellite loci were obtained had an advantageous effect on growth traits and these loci were as the reference markers for marker~assisted breeding of Songpu mirror carp.A pair of specific primers was designed according to common carp MHC II B geng sequences on NCBI. In this study, the primer was used to analyse the uniform fragment of 108bp of the exon 3 of MHC II B gene by PCR~SSCP in six common carp population. Result showed: Seven genotypes (A~G) were acquired in the population. The most common genotype was genotype B that presented in all population and accounted for 47.45%. The ratio of other genotypes varied from 1.1% to17.15%. Four variable nucleotide positions and three variable amino acid positions were detected after fragments of different genotypes were recycled and sequenced. The populations of Yibu common carp and Big~head common carp had four(3.7%) variable nucleotide positions.Three(2.7%) variable nucleotide positions were found in the populations of Songhe common carp, Songpu common carp and German mirror common carp. The population of Songpu mirror common carp had only two (1.8%) variable nucleotide positions. Oberved heterozygosity(Ho) , expected heterozygosity(He) and polymorphism information contents (PIC ) were determined. The observed heterozygosity value (Ho) was from 0.1037 to 0.7464, the expected heterozygosity value (He) was from 0.0652 to 0.4825 and the PIC value was from 0.0625 to 0.3609. These date provide great help for the study of the polymorphisms of MHC gene and for the selected breeding of disease resistance in common carp.
