Povidone-iodine(PVP-I), as efficient disinfectant, widely used in aquaculture disinfection. However, with the rapid development of aquaculture, aquatic drugs extensive use of extensive and far-reaching impact on the aquatic environment, the environmental ecological problems resulting increasing people’s attention. In recent years, povidone-iodine to crabs, fish, shellfish, and shrimp etc. aquatic animals acute toxicity studies have been carried out, as well as the research of cytotoxic mechanism at home and abroad. From the existing literature, povidone-iodine in safety evaluation of the aquaculture environment rarely has been reported, resulting in there is a big blindness and one-sidedness of the aquaculture environment safety evaluation. In addition, the study found that povidone-iodine safe concentration of the different fingerling are quite different, the concentration in the killing of aquatic pathogens are not the same. Therefore, the strengthening povidone-iodine security is essential to promote the reasonable application process in aquaculture. The entire thesis can be divided into three parts:1Carried out povidone-iodine on the acute toxicity studies of zebrafish and five common aquaculture animals.Experiment with reference to "the GB/T13267-91Water quality-Determination of the acute toxicity of substance to a freshwater fish (Brachydanio rerio Hamilton-Buchannan)" The results showed PVP-I to zebrafish96h-LD50was17.63mg/L, the safety concentration was1.76mg/L; to Carassius auratus gibelio96h-LD50was74.77mg/L, the safe concentration was7.48mg/L; containing to Ctenopharyngodon Idellus96h LD50was64.69mg/L, the safe concentration was6.47mg/L; to Procambarus clarkii96h-LD50was13.89mg/L, the safe concentration was1.39mg/L; to Penaeus vannamei Boone96h-LD50was7.781mg/L, the safe concentration was0.78mg/L; to tilapia96h-LD50was33.25mg/L, the safe concentration was3.33mg/L. 2Carry out povidone-iodine ecotoxicological studies. with reference to"GB/T21805-2008Chemicals-Alga growth inhibition test""the GB/T13266-91Water quality-Determination of the acute toxicity of substance to Daphnia(Daphnia magna straus) Fishery drugs in clinical trials technical specifications(Bureau of vet of pasturage of Ministry of Agriculture2003)", the growth inhibition of the PVP-Ⅰ on Chlorella sp., its inhibitory effect on aquatic probiotic bacteria and its acute toxicity to Daphnia magna straus were observed, its influences on main harmful physicochemical factors of its farming water were also assayed. The results showed that the growth of Chlorella sp. was promoted with PVP-I at the final concentrations of6.00~14.00mg/L, its minimum inhibitory concentration to aquatic probiotics were ranging from128mg/L to512mg/L, its50%inhibitory concentration to Chlorella sp. was estimated to be above14.00mg/L, and its50%lethal dose to D. magna straus, B. rerio were13.44mg/L,17.63mg/L,74.77mg/L. In addition, and in the period of14days after the adding of the PVP-Ⅰ into the farming water at0.20-1.40mg/L, the contents of the ammonia and nitrite in farming water were gradually reduced with the increased concentration of the PVP-Ⅰ.3Experiment using a test double broth dilution method, is about to PVP-Ⅰ were added to a test tube containing2ml of sterile nutrient broth, to a final concentration:0mg/L、0.125mg/L、0.25mg/L、0.5mg/L、1mg/L、2mg/L、4mg/L,8mg/L、16mg/L32mg/L、64mg/L、128mg/L、256mg/L、512mg/L、1024mg/L、2048mg/L、4096mg/L, and a negative control group,scilicet blank sterile nutrient broth group,Strains were inoculated with the bacterial suspension to a final concentration of3.0×108cfu/ml after incubation at28℃for24h observation. Sterile growth, dilution of povidone-iodine concentration regard as (MIC) of the experimental strains.0.1mL were taken from each tube of all bacterial growth, was applied in the appropriate sterile agar plates, placed in a constant temperature of28℃in the incubator to culture for24h and observe the results.The concentration of the the sterile growth plate the minimum quality is the minimum bactericidal concentration of quality.The results showed that: povidone-iodine to Yersinia Ruckeri sp.、Citrobacter freundii sp. strains、Aeromonas hydrophila sp.、Vibrio Parahaemolyticus sp、Vibrio alginolyticus sp. bacteriostatic and bactericidal effect is not satisfactory, povidone-iodine to Vibrio harveyi sp、 P.Aeruginosa sp. bacteriostatic and bactericidal effect is satisfactory.
