The paper described the antigiardial activity of osthole in vitro and in vivo, andprovided a theoretical basis for the application of osthole in prevention and treatmentof canine giardiasis.The effects of osthole on giardia canis in vitro To observe the antigiardialactivity of osthole,we firstly analyzed the effect of the different concentrations ostholon growth inhibition of trophozoites, and then we described that the effect of ostholeon cell viability assays at IC50concentration.The growth inhibition assay showed thatall the different concentrations of osthole have a certain of growth inhibition.As theconcentration of osthole increased, the more obvious growth inhibition,especially the78.62%reduction was observed at concentrations5.0mg/mL. The viability assaysrevealed that osthole inhibited significantly the cell viability using its IC50concentrations.Effect of osthol on Ultrastructure of giardia canis in vitro To study themechanism of anti-giardial activity of osthol, the trophozoites were morphologicallyobserved respectively with scanning electron microscopy and transmission electronmicroscopy after treated with the drug. The results showed that, after treated by theosthol, the trophozoites have obvious lesions in the surface of ventral sucker,membrane rupture of the ventral sucker, cell contents leakage, lamella structure and alot of vacuoles in the cytoplasm. and the nuclear became deformed and chromatinmargination,and the disintegrate of the discs was seen.The effects of osthole on giardia canis in vivo Twenty-four dogs were infectedwith Giardia canist rophozoites and observed the therapeutic effect of osthole on dogsinfected with Giardia canis. The results showed that14dogs were successfullyinfected with Giardia canis among22dogs. The high dose of osthole（100mg/kg）had the best efficacy with significantly reduced the number of cysts and shorten theduration,and the therapeutic effect was a dose-dependent of osthole.
