| [Object] This study aimed at the hybrid offsprings all male that is female offsprings all deaths during embryonic development, as the estrogen receptor gene (ER) and androgen receptor (AR) gene candidate genes in chicken, Using quantitative PCR to detection AR, ER gene expression levels in different gender, different time points on the basis of sexes identification.Comparative study to investigate the AR, ER gene expression to the mechanism of action of early death in chicken, quail and its hybrids.[Method] Insemination for chickens (♂)×quail (♀), the three groups of egg were incubated simultaneously. The embryo of age60h,72h,84h,96h,108h,120h,132h,144h,156h,168h total of10time points embryonic samples were collected and statistical fertilization rate. Genomic DNA was extracted by the method of CHD gene-specific amplification to sexes identification. Five time points were selected and male as well as female individuals embryonic, RNA was extracted and using real-time PCR to detect the AR gene and the ER gene level expressing significant difference test, using statistical methods and analysis and hybrid poultry associated with embryonic development.[Conclusion]1. Artificial insemination and fertilization rate statisticscollected2605chicken (♂)×quail (♀)eggs, the highest fertilization rate was47.37%, the lowest fertilization rate was22.5%, the average fertilization rate was32.25%, live embryo rate was42.35%; collected152chicken (♂)×chickens (♀) embryos,146live embryos, the fertilization rate was96%; collected quail (♂)×quail (♀)62embryos,59live embryos, fertilization rate was95%.2.The sex ratio of chickens, quail, and hybrids poultry in different developmental timeTo identify the unknown sex embryos of Chickens142, quails57and hybrids153, the male and female ratio of Chicken is0.90:1; quail is0.84:1. The hybrids is0.63:1, and the difference was significant.3.Chicken, quail, and their hybrids avian embryonic AR, ER expression differences and developmental changes.The expression patterns of AR, ER gene in chicken and quail72-168h early embryonic tissue, respectively, were similar, but the expression patterns of hybrids is different during this time,indicating that the AR and ER genes showing abnormal at72-168h in hybrid embryos.These results suggest that the abnormal expression of AR, ER gene in hybrids early embryonic at72-168h may cause death of a large number of hybrid embryos.[Conclusion] the abnormal expression of the ER and AR gene may be a major cause of death caused by early hybrid embryos in chicken and quail early hybrids. |
