microRNA play an important role in the process of growth and development of ovarian, ovum, early embryo and other tissues and organs that are related to reproductive performance, but studies about the regulation of microRNA in goats’reproductive performance have not been reported. This experiment test the lambing mechanism from the transcriptome level.This experiment provides experimental basis for increasing the lambing rate of dairy goats by constructing the microRNA bank of estrus ovarian tissue of both polytocous and monotocous dairy goats, using high-throughput sequencing technology to screen differential expression, applying bioinformatics to analyze gene structure and potential function, and identifying microRNA related to sheep lamb traits. The main conclusions are:1. According to the sequenced results of the three admixed ovarian sample, which combined both low litter size dairy goats (HBZ01) and high litter size dairy goats (HBZ02), there are7859386pair of original data and4911179pair of (127205types) microRNA sequences resulting from low litter size dairy goats,7859386pair of original data and28941514pair of (454911types) microRNA sequences resulting from high litter size dairy goats.2. From the comparison of microRNA length, there are2018266pair of microRNA sequences at the length of22nt, occupying41.10%of the total in HBZ01. In HBZ02, there are8856575pair of microRNA sequences at the same length, occupying39.26%of the total. There also shows a phenomenon of22nt enrichment in the of miRNA species statistics.3. Using quantitative real-time PCR technology to detect6kinds of differentially expressed miRNA (PC-5p-5964110, bta-miR-181cR-2, bta-miR-425-5pL+1, bta-miR-10bR-1, eca-miR-34b-3p2ss9AG10CT, bta-miR-223), the results correspond to those on the construction of the built bank, showing that the quality is conformance to specification.4. In accordance with the requirements for secondary stem loop structure, this study predicts241types of new microRNA. From the point of expression level, PC-5p-5964110belongs to high expression.5. There are630types of differential expression microRNA between high low litter size dairy goats, including newfound31types. From the detected analysis statistics of the630 types of differential expression microRNA:487types of microRNA co-express in two goats’ ovarian tissues, only143types of microRNA express in polytocous goats’ovarian tissue. Cluster analysis demonstates that t hese are two microRNA (ssc-miR-451R-1and bta-miR-486R-1) in HBZ02that are extremely significant different when compared with that in HBZO1.
