A method for determination of the OTC concentration in yellow catfish plasma and other tissues by High Performance Liquid Chromatography was established in this study. Based on this method, the effect of temperature and time administer drug on OTC pharmacokinetics in plasma and tissues of yellow catfish (Pelteobagrus fulbvidraco), and the residue and depletion of OTC in yellow catfish. The primary result of this study was listed as follows.A method for determination of the OTC concentration in yellow catfish plasma and other tissues by HPLC was established. Chromatographic condition:Agilent SB-C18(250mm×4.6mm i. d.,5μm) reversed-phase column was used, acetonitrile NaH2PO4+EDTA(V:V=22:78) as the mobile phase, ultraviolet wavelength was355nm, column temperature was25℃, flow rate was0.5mL/min. The results demonstrated that the concentration in the range form0.5μg/mL to20.0μg/mL, the concentration had a strict linear relationship with peak area; the standard recovery rates of tissues were between78.7%-98.6%; each of the relative standard deviation (RSD) was less than8.8%; the limit of quantification of OTC in yellow catfish tissues were0.03μg/mL,0.05μg/g,0.04μg/g,0.05μg/g for plasma, muscle, liver and kidney, respectively.The pharmacokinetics studies of OTC in yellow catfish following a single oral administration with the dose of100mg/(kg-bw) were conducted at18℃and25℃. And the pharmacokinetics study of OTC in yellow catfish following a muti-dose oral administration with the dose of100mg/(kg-bw) for5days was conducted at25℃. The results showed that a one-compartment model was developed to describe the pharmacokinetics of OTC in yellow catfish. At25℃after the single dose, the half-life (t1/2(ka)) of absorption was3.92h,1.44h,2.75h, and3.34h in plasma, muscle, liver, and kidney, respectively; the half-life (ti/2(ke)) of elimination was7.64h,19.08h,26.29h and21.44h in plasma, muscle, liver, and kidney, respectively; the order of peak concentration (Cmax) were liver>kidney>plasma>muscle, they were3.48μg/g,2.90μg/g,1.46μg/mL, and1.39μg/g. At18℃after the single dose, the half-life (t1/2(ka)) of absorption was3.92h,2.66h,3.65h and3.25h in plasma, muscle, liver, and kidney, respectively; the half-life (t1/2(ke)) of elimination was8.98h,29.60h,21.16h and21.04h in plasma, muscle, liver, and kidney, respectively; the peak concentrations (Cmax) were1.28μg/mL,3.33μg/g,2.53p.g/g and1.34μg/g in plasma, muscle, liver, and kidney, respectively. At25℃after muti-dose, the half-life (t1/2(ka)) of absorption was0.35h,0.42h,0.32h and0.22h in plasma, muscle, liver, and kidney, respectively; the half-life (t1/2(ke)) of elimination was47.54h,49.87h,47.73h and70.99h in plasma, muscle, liver, and kidney, respectively; the peak concentrations (Cmax) were4.17μg/mL,14.02μg/g,8.51μg/g and3.84μg/g in plasma, muscle, liver, and kidney, respectively. The results suggested that the absorption of OTC in yellow catfish was limited. The effect of temperature on absorption of OTC in yellow catfish was really low, however, temperature has a great influence on elimination rate constant (Ke). OTC was absorbed faster after the multi-dose than after the single dose. Whereas, OTC was discharged slower from yellow catfish compared to the single dose and with a higher peak concentration (Cmax). It suggested the accumulation of OTC in yellow catfish.In order to study the elimination of OTC in yellow catfish muscle, at25℃, following oral administration at a dose of100mg/kg of OTC per day for5days, the fish were sampled at certain time, and the concentration were determined by HPLC. The average recovery rate of OTC in yellow catfish muscle was80.2%, and the limit of detection was0.05μg/g. The results revealed that the elimination of OTC from muscle of yellow catfish was very slow, the withdrawal period of OTC in the muscle of yellow catfish was46days until the concentration of OTC reducing to0.1μg/g under this experiment condition.
