Intestinal mucosal immune system is the body’s largest and most complex parts of the immune system.For newborn animals, the development of the intestinal mucosal immune system seems to be largely relieson the microbial exposure. Bacteria stimulate the body to produce a lot of lymphocyte and lymphoid tissue,promoting the mucosal immune system of the normal development and mature. In this experiment,newborn lambs were supplemented with four kinds of ruminal fluid preparations, and observed the effect ofpreparations on the intestinal mucosal immunity of the lambs.Experiment Ⅰ Macrophages RAW264.7and the mouse whole blood as the model, different treatmentof ruminal fluids was used for cell culture in vitro, results showed that ruminal fluid involved immuneactive substances, which could stimulate the macrophage to produce the TNF alpha, and RAW246.7macrophage was more confortable for this experiment.Experiment Ⅱ Newborn lambs as model animals, were fed with healthy adult sheep ruminal fluid andits components (ruminal fluid supernatant, autoclaved ruminal fluid supernatant, ultrasonic processingrumen fluid supernatant, autoclaved ultrasonic processing rumen fluid supernatant) at1-day-old.Slaughtered three lambs of each group for receiving the alimentary tract, and determinated the content ofIgA, SIgA and IgG in small intestinal mucsal protein of24-old-day. Results showed that the content of IgA,SIgA and IgG in small intestinenal mucosal protein in all trials was higher than the control group. The totalcontent of IgA in the experimental groups was increased by24.68%(P>0.05),15.84%(P>0.05),28.56%(P>0.05)and9.26%(P>0.05), respectively. The total content of SIgAin trial groups was increased by7.14%(P>0.05),25.32%(P>0.05),27.60%(P>0.05)and14.28%(P>0.05), respectively. The contentof IgG in each trial group was increased by4.71%(P>0.05),6.99%(P>0.05),37.18%(P>0.05) and36.39%(P>0.05), respectively. In general, the content of immunoglobulins in ileum was higher thanduodenum and jejunum, the total immunoglobulin in trial groups was higher than the control group. Theconcentration of immunoglobulin in plasma at28-day-old was higher than14-day-old lambs, in which theconcentration of IgG increased significantly.This present study showed that the adult sheep ruminal fluid preparations involved immune activesubstances, which can stimulated the macrophage and mouse whole blood to produce the TNF alpha.RAW246.7macrophage was more confortable than whole blood of mouse. Newborn lambs weresupplemented with ultrasonic processing rumen fluid supernatant, which could significantly improve thecontent of IgA in the small intestinal mucosal protein of24-day-old lambs, but has no significant influenceon the content of SIgA and IgG in the small intestinal mucosal protein of24-day-old lambs. Theconcentration of immunoglobulins of the plasma was not signigicantly affected by supplementation withfour kinds of ruminal fluid preparations of lambs.
