This paper to the actinomycetes separation identification in Baicheng yanshan.the freeculture method,Separated and cultivated, based on16S rDNA gene sequence systemdevelopment analysis in the baicheng yanshan the diversity of actinomycetes, so as to getthe yanshan actinomycetes species resources. The follow that free training results as theresearch foundation, through the can cultivate actinomycetes separation and training,more accurate for xinjiang the baicheng yanshan actinomycetes resources, to explore newactinomycetes species to agricultural production harm serious plant common pathogenicbacteria for target bacteria for good antagonistic radiate strain screening for thedevelopment and utilization of extreme environment to provide technical support andactinomycetes strains support. Get results in the paper are as follows.(1)Use of actinomycetes specific primer amplification16S rDNA gene, constructing the16S rDNA gene cloning library, the library clonal fragments RFLP (Restriction FragmentLength Polymorphism, Restriction Fragment Length Polymorphism) analysis. Theanalysis shows that the216clone distribution in actinomycetes steel(Actinobacteria) threesubclass, namely actinomycetes subclass (Actinobacteridae171),79.2%of the totalnumber of cloning, acid micro bacteria subclass (Acidimicrobidae45) to total cloningnumber of20.8%, and in GenBank known micrococcus sequence maximum similarity isless than92%, is the acid micro bacteria subclass new groups. Advantage actinomycetesactinomycosis subclass bar bacillus suborder nocardia division (Nocardiaceae)Rhodococcus (Rhodococcus), accounting for51.9%of the total number of cloning.24.5%of the sequence and has published effective strains of sequence similarity is less than97%,in part, sequence formed several independent evolution branch, may represent aactinomycetes new groups.(2)There are35strains of separation actinomycetes by used different salt concentrationof9kinds of selective medium, which distribution in actinomycetes purpose4suborder:chain sporocyst bacteria suborder (Streptosporangineae) of the chain single spore bacteriagenera (Streptomonospora)4strains; Quasi nocardia (Nocardiopsis)17strains,accounting for48.3%of the total number of bacterial strain separation; salt radiate sporebacteria genera (Haloactinospora)7strains; Saccharopolyspora4strains; Actinomycetesmany spore bacterium and objective radiate more spore bacterium is7strains. Four strainactinomycetes16SrDNA gene sequence and has published the largest effective strainssequence similarity is less than97%, as potential actinomycetes new classification unit.(3) The results show that there are three strains of actinomycetes have to cottonyellow blight of antagonistic activityand Phytophthora capsici very good. in yanshanseparated from the35strains actinomycetes are antagonistic actinomycetes screening.
