Abstract/Summary:
In order to study the fall blood sugar factor of the banana flower, ba west banana flower of hainan province as object,using the qualitative method to analysis active ingredients of banana flower, and the extraction process of the total flavonoids with the blood sugar reduction factor in banana flower was optimized. For banana flower extract further made safety evaluation on the acute toxicity test in mice. With alpha glycosidase enzyme inhibition activity screening method,banana flowe extraction, isolation, purification and structure identification of the compounds was reviewed.1The basic experiment of banana flowerIn this paper qualitative analysisr the effective components from the banana flower, the results showed that the banana flower contains fall blood sugar activity function factor, such as saponins, terpenes, flavonoids, alkaloids, etc. In order to research optimum parameters of extraction of the flavonoids in banana flower, ethanol as solvent,rutin as referencethe,with the extration ratio of flavonoids as indicators, using heat reflux extraction. On the basis of single factor experiment, the choice of ethanol concentration, solid-liquid ratio, extraction temperature, extraction time by orthogonal experiment,with ethanol reflux extraction flavonoids in banana flowers results show that the best optimum process:60%ethanol concentration, solid-liquid ratio of1:25and at extraction temperature65℃, the extraction time of1h. Evaluation for banana flower extracts on acute toxicity experiment, the results obtained:banana flower extracts of KM mice to dose of5g/kg, delivery volume is0.4mL/10g. After the treatment and recovery observating did not see animals appear the symptom of abnomal reaction, the test group, negative control group and blank control group on average weight gain is normal, after autopsied did not see unusual in the major organs, explain banana flower extract is safe.2Alpha Glucosidase activity of banana flower extractPreliminary investigation on banana flower extract with alpha glycosidase enzymes activity analysis, and alpha Glucosidase inhibitory type were determined. Acarbose as positive control, the determination of banana flower extract alpha Glucosidase activity inhibition rate, the results show that under the same concentration, banana flower extracts on alpha Glucosidase activity has a different degree of inhibition, the alpha Glucosidase to banana flower ethanol extracts have stronger inhibition of enzyme activity, inhibition rate can reach88.56%. Alpha Glucosidase inhibitors inhibit type is competitive inhibition, and alpha GlucosidaseKm=674.074ug/ml; Banana flower petroleum ether part of alpha Glucosidase inhibitory effect than acarbose, enzyme inhibition rate is as high as73.98%, IC50was788.36mg/ml; Extract the highest part of alpha Glucosidase inhibitory activity type is competitive, secondly ethyl acetate part of inhibition rate was52.78%, the IC50was1877.77mg/ml, under the same concentration of acarbose inhibition rate was56.45%, the IC50was999.31mg/ml. On the components of volatile oil part with petroleum ether from banana flower with GC-MS analysis, the results detected31species composition,29compounds were identified, main chemical components of organic acids (71.58%), esters (13.01%), amine (5.88%), aldehyde (1.52%), ketone (0.42%) of compound. Content in more than1%of the ingredients are hexadecanoic acid23.40%, linoleic acid27.29%and13.73%oleic acid, phthalic acid butyl ester5.83%, stearic acid3.67%, diphenylamine3.50%, hexadecanoic acid ethyl ester was3.17%,1.44%linolenic acid ethyl ester,1.25%vanillin and hexadecanoic acid methyl ester1.24%, different Ding Jilin phthalate (1.05%).3Banana flower extraction and separation, purification and structural identification of compoundsThis study that extracted the four compounds using the many kinds of column chromatography separation and purification from banana flower ethanol extracts the five compounds, according to the physical and chemical properties and spectroscopic data were identified as:vanillic acid (1), ferulic acid (2), beta sitosterol,(3) beta carrot glycosides (4),2-mehtoxy-9-(4’hydroxyphenyl)-phenalen-1-one (5) all of these are for the first time isolated from banana flower. Of the five compounds and active screening of IC50alue concentration,1(IC50=2004.58ug/mL),2(IC50=1258.35ug/mL),3(IC50=283.67ug/mL),4(IC50=247.35ug/mL),5(IC50=3.86ug/mL); By doing Lineweaver Burk curve of compounds3and4is a competitive inhibition, compound5is mixed inhibition, and concluded that compound3Ki=20.09ug/ml, compound4Ki=2.34ug/ml, compound5Ki=4.40ug/ml.
