Cordyceps sinensis is a traditional Chinese medicine which has a significant role in keeping healthy and preventing disease.Howerver the lack of natural Cordyceps leads the market in short supply.Therefore, the Cordyceps artificial fermentation technology has been attented.Whether the mycelium produced by different fermentation process have the same pharmacological effects with the natural Cordyceps has becomed a hot spot of research in this field.In this study, we use the Cordyceps mycelium powder which produced by the solid-state fermentation as the experimental materials stated. We extract and purify a single-component Cordyceps polysaccharide(CSPS), preliminary inquiry the composition of the polysaccharide, study the activity of increasing immunization, anti-tumor and anti-oxidant. The major fingdings were listed as follows:1. A single-component CSPS was isolated and purfied from the Cordyceps mycelium. The molecular weight of CSPS were about1.17×105detected by Gel filtration method. Fourier transform infrared spectrometer analysis showed that CSPS contains characteristic absorption peak of polysaccharide and pyran ring. According to GC analysis, CSPS was composed of rhamnose, xylose, mannose, glucose and galactose, The composition ratio was1:2.39:5.40:30.67:13.37. Results of periodate oxidation and Smith degradation suggested that CSPS containing1â†'2,1â†'3,1â†'4,1â†'6glycosidic bond.2.The result of immunological activity in vitro show that:the CSPS can enhance the phagocytosis vitality of mouse peritoneal macrophage, promote peritoneal macrophages release NO and the proliferation of spleen cell and the production of tumor necrosis factor (TNF-α) in splenocytes, which have a certain concentration dependent effect.3.The potential of CSPS in inhibiting S180cells viability was assessed by MTT assay. CSPS has a certain inhibition ability in cell proliferation with a significant dose-dependent. Early apoptosis was dectected by flow cytometry using Annexin V-FITC and PI staining, the experiment also shows the same conclusion. 4. CSPS reducing power test results show that it has certain reducding ability. Hydroxyl radical scavenging experimental results show that CSPS has a certain ability of scavenging hydroxyl radical.Superoxide radical scavenging experimental results show that CSPS has a certain ability of superoxide free radical. As the polysaccharide concentration increased, the reducing power and the clearance rate is rising.
