Abstract/Summary:
The fresh seed often show late ripe of physiology after picking up. Using silicone drying method at room temperature,the decrease of moisture content in the new picking seeds of Celtis sinensis from10.74%to6.30%、5.55%、4.03%and3.60%。Researching every index of physiology and biochemistry after restoring1、2、3month under the room temperature and4centigrade.At the same time,study the germination of the moisture content of10.74%after seed soaking.The concrete results were as following:1、The specific biology characteristic of seeds of Celtis sinensis were as following:the clarity of the seed lot was96.43%, the vigour of seeds was32%.The structure of seeds’ embryo was complete and the seed coat was about70.87%of the seeds’ weight,the embryo was about29.13%.The1000-seed weight was47.456g so that it belonged to the medium-sized seed.The relative moisture content of seeds was10.74%,the seeds reached saturation afer water uptaking for48hours and the saturation moisture content was48%,so the seed coat had the phenomenon of watertightness.2、The seeds of different moisture content restored under the normal temperature and4centigrade for1、2、3month have differrnt soluble sugar content.The change trend showed that high moisture content was high, low moisture content was low, and it increased along with the storage time.The moisture content of5.55%was useful to break the seed dormancy under the sturage of4centigrade for3months.During the storage processing,the change of soluble protein content of the same seed moisture content was not significant. After storing1month,the soluble protein content of all seeds had increased significantly.Among all the seeds,the moisture content of5.55%seeds’ soluble protein content showed the tend of low-high-low,and the difference was significant,the storage effect was the best.3、During the different stage of storage processing,the content of proline was higher,but the change trend was not obvious.The higher moisture content seeds needed to produce more proline to resist external disadvantage factores during the temporary storage. The moisture content of5.55%seeds were useful to store under4centigrade.But the3.60%seeds stored under4centigrade for1month had the best storage effect.The content of malondialdehyde of the fresh seeds had2maximum during the storage,the seeds of6.30%appear serious membranous peroxide phenomenon at the beginning of the dehydration.At the sroage of2months,the malondialdehyde conten of seeds of3.60%、4.03%were all over0.300μmol/L.The5.55%seeds had the best storage effect under4centigrade.4、The activity of Superoxide dismutase and Polyphenol oxidase under the storage of4centigrade were better than the room temperature,among all the moisture content,the5.55%seeds had relatively stable activity during different stages,and present the change trend of low-high-low,the activity of enzyme reached the maxmium and to drop off over the storage time.Moisture content of seeds was too high or too low will cause the activity of enzyme changes mussily without regularity.The activity of Peroxidase appeared the phenomenon of inactivation during the storage processing.The3.60%seeds stored2months and the5.55%store1month under the room temperature all had this phenomenon.The activity of enzyme of all seeds under4centigrade were normal,but the variation was not obvious.The activity of5.55%moisture content seeds keep the level of relative stability.The change trend of the activity of Catalase appered low-high-low,and reached the maxmium after2months under the room temperature,the activity of enzym dropped off along with time.The activity of enzyme stored under the storage of4centigrade were all better than the room temperature.5、The seeds couldn’t germinate in different exogenous reagent seed soaking.At the same time,seeds produced different degree of mildew rate.During all the processing, the electrical conductivity was the lowest for the seeds with the treatment of PEG but with the highest mildew rate.The seeds soaking of50mg·L-1GA3、100mg·L-1NAA、200mg·L-16-BA and200mg·L-1IAA have the lowest mildew rate.Using the seeds under different storage processing to do the germination experiment,all the seeds couldn’t germinate,the5.55%seeds store2months under4centigrade have the lowest rate.
