Bupleurum chinense DC. is a well-known traditional chinese medicine, which belongs to Bupleurum L. of Umnelliferae,and was recently named as the original plant of authentic herbs by People’s Republic of China Pharmacopoeia. Its root contains saikosaponins as the main active components, which have antipyretic, sedative, analgesic, antitussive, anti-inflammatory, anti-virus, hepatoprotective and anti-tumor activities.The conditions of using B. chinense adventitious root system to produce saikosaponins efficient were studied by weans of adding elicitor and precursors.The results showed that the contents of saikosaponin were affected by amino acids, elicitor and precursors. The1/2MS medium combined0.5mmol/L tyrosine,0.5mmol/L lysine,1.0mmol/L threonine,1.0mg/L double acid calcium diphenylurea and50g/L sucrose was the most suitable medium for B. chinense from Ruicheng. The1/2MS medium combined0.1mmol/L tyrosine,0.1mmol/L lysine and0.1mmol/L threonine was the most suitable medium for B. chinense from Yangqu. The1/2MS medium combined0.1mmol/L tyrosine,1.0mmol/L lysine and1.0mmol/L threonine was the most suitable medium for B. chinense from Zuoquan. The1/2MS medium combined0.5mmol/L glutamine,1.0mmol/L aspartic acid and0.1mmol/L aspartic acid was the most suitable medium for B. chinense from Ruicheng. The1/2MS medium combined1.0mmol/L glutamine,1.0mmol/L aspartic acid and0.5mmol/L aspartic acid was the most suitable medium for B. chinense from Yangqu. The1/2MS medium combined1.0mmol/L glutamine,0.5mmol/L aspartic acid and0.1mmol/L aspartic acid was the most suitable medium for B. chinense from Dingxiang. The medium combined1.0mmol/L phenylalanine was the most suitable medium for B. chinense from Wanrong and Ruicheng. The medium combined1.0mmol/L alanine was the most suitable medium for B. chinense from Ruicheng. The medium combined0.5mmol/L alanine was the most suitable medium for B. chinense from Wanrong.The medium combined111.2mg/L Fe2+,17.2mg/L Zn2+and89.2mg/L Mn2+was the most suitable medium for B. chinense from Ruicheng. The medium combined55.6mg/L Fe2+,34.4mg/L Zn2+and89.2mg/L Mn2+was the most suitable medium for B. chinense from Dingxiang.
