The paper is divided into two parts:1) Fine mapping of a control gene LBVP(t) in rice growing period;2) Functional analysis of a rice zinc-finger gene ZOS5-07.Rice heading time is mainly decided by the photoperiod sensitivity and basic vegetative growth. Our laboratory found a mutant lbvp (long basic vegetative growth period) from mutagenic Nipponbare induced by x-ray with a long basic vegetative growth period, which is decided by a single recessive gene, the gene has been temporarily named as LBVP (t). We used an F2population constructed by crossing lbvp mutant with an indica cultivar Acc8558to map the gene on the basic of primary mapping.23new SSR markers and23Indel markers were designed according to the rice genome sequence, from which4pairs of SSR markers and5pairs of Indel makers showing polymorphism between the two parents were obtained. By linkage analyzing the F2mutants with these9markers, LBVP(t) was finally mapped in a region of0.9cM between two PAC clones on chromosome6, the physical distance is180Kb. There are23candidate genes in this region, probably identified the gene as an O-methyltransferase gene or a ring zinc finger gene. Gene sequencing and expression analysis will be done.Root is an important nutrition organ of rice, it has roles of fixation, absorbing nutrients and moisture. Our laboratory found a gene ZOS5-07preferentially expressed in rice root when analysing zinc finger genes expression chip data, it encodes a C2H2zinc-finger protein which probably involved in the development of rice root. In the study, an overexpression vector Ubi:ZOS5-07, a vector pTCK-ZOS5-07of RNA intervention and a GUS expression test vector pZOS5-07::GUS have been constructed, they have been transferred into japonica cultivar rice zhonghual5. We obtained18To populations with ZOS5-07over expressing and21T1populations with RNA interfered of ZOS5-07, but the GUS transgenic plants are on differentiation culture medium. By analyzing the root morphology of the seedlings, enhancing the expression of ZOS5-07, the taproot are longer than wild-type, and they have more lateral roots.while the RNA intervention has an opposite result. The result indicates that ZOS5-07may participate in regulating the root development of rice.
