| Sulfachlorpyrazine (SPZ) is an effectively wide spectrum antimicrobial, belonging to the sulphonamides. SPZ is widely used for the treatment of various poultry diseases, particularly coccidiosis, and also be used for treatment fowl typhoid and fowl cholera when coccidian breaks out. With the wide use of SPZ, coccidian have acquired high drug resistance. Diaveridine (DVD) is an antisepsis synergist for animal, the application of the combining DVD and SPZ can enhance SPZ antibacterial activity and displays bactericidal action, the combination also results in fewer resistant bacterial strains. Study on the elimination regularity of SPZ-DVD solution residue in broiler tissues is in order to establish a withdrawal time,so as to provide references for SPZ-DVD solution rational use in the clinic.1. Determination of SPZ and DVD residues in broiler tissuesSPZ was extracted with acetonitrile and acetone. Defatted with n-hexane and cleaned up with MCX solid-phase extraction. The mobile phase was acetonitrile-0.017mol/L phosphoric acid (32/68, v/v). A method of HPLC with ultraviolet detection was applied, quantified by external standard. The results showed that the concentration of SPZ had a good linear coefficient relation (R2=0.9999) in the range of0.04-8mg/L. The limit of detection was0.02mg/kg,the limit of quantitation was0.04mg/kg.The results showed that the mean recoveries of SPZ spiked in broiler tissues at three concentrations of0.04,1.0and8.0mg/kg were between84.56%～95.10%,the within-day CV and between-day CV were1.88%～8.39%and1.10%～7.88%, respectively. The established method can be used for residue analysis of SPZ in broiler tissues.DVD was extracted with acetonitrile and acetone. The results showed that the mean recoveries of DVD spiked in broiler tissues at three concentrations of0.05,1.0and8.0mg/kg were between83.83%～94.05%,the within-day CV and between-day CV were2.37%～7.46%and1.33%～6.07%, respectively. The established method can be used for residue analysis of DVD in broiler tissues.2. Study on the residue elimination of SPZ-DVD Solution in broiler36healthy broilers were administered SPZ-DVD solution per day at the highest dosage for seven successive days. Six broilers per day were slaughtered at0,1,3,5,7,9days after the last administration. SPZ and DVD in broiler tissues were determinted by an established method above. The results showed the concentrations of SPZ in all tissues were was (2.629±2.066)μg/g, (0.831±0.694)μg/g,(0.364±0.241)μg/g,(0.328±0.316)μg/g for kindy, skin+fat, liver, muscle at day0, respectively. The concentrations of DVD in all tissues were was (1.995±0.625)μg/g,(0.266±0.135)μg/g,(0.810±0.523)μg/g,(0.196±0.105)μg/g for kindy, skin+fat, liver, muscle at day0, respectively.SPZ were quickly eliminated in liver and muscle, and slowly eliminated in kindy and skin+fat.This shows that the kindy and skin+fat are target organs of SPZ. DVD were quickly eliminated in liver, and slowly eliminated in kindy. This shows the kindy are target organs of DVD. The time when the concentrations of SPZ in muscle, liver, kindy and skin+fat were lower than MRLs was3d、3d、5d、5d and the DVD was3d、1d、7d、5d. According to the residues of SPZ、DVD in broiler tissues and the rule of MRL. And the raw data were analysed with WT1.4program.The withdrawal period was8.93d and8.82d in skin+fat and kindy for SPZ, respectively.The withdrawal period was7.79days in kindy for DVD. Results of this experiment showed that the withdrawal period of SPZ-DVD Solution in broiler was9days. |
