To find the genetic resources of pre-harvest sprouting resistance in Sichuan wheat accessions and know the genetic variation of Vp-1, eleven specific PCR primer sets were used to detect allelic variation in the Vp-1gene in the landrace and cultivar accessions. Meanwhile, a total of86accessions (40landraces and46cultivars) were also used to carry out dormancy tests and in a correlation analysis between Vp-1variation and dormancy level. The main results and conclusions are summarized as following:1. Sichuan wheat landraces and cultivars had a wide range of dormancy levels. The average germination index (GI) of Sichuan landraces was0.232, whereas it was much lower for cultivars (0.834). The result indicated that Sichuan wheat landraces were more resistance to PHS, compared to cultivar. The average dormancy levels of wheat accessions with different grain colors were different, and the GI of white grained accessions (0.878) was2fold higher than red grained accessions (0.443), indicating white grained wheat was more susceptible to PHS than red grained wheat.2. Phylogenetic distances of the accessions were calculated and used to construct a neighbor-joining tree based on the Vp-IB alleles. The accessions clearly divided into three groups. Phylogenetic analysis therefore indicated that the diversity of the Vp-1B was related to the cultivar and landrace groups. The39PCR fragments were numbered for analysis in86accessions (carried out dormancy tests), indicating some PCR fragments were specific to landraces or cultivars and to white or red grained accessions.3. Based on Vp-1B3marker and GI, nine PHS resistance accessions (87-429, Xifu1, Anyuenuermai,Guanxian Baihuaganerzimai, Langzhong Fangmai, Luxian Guangtouhonghuamai, Luxian Guangtoumai, Nanchong Sanyueheshangmai,and Zhigong Yicengmai) were screened out.4. Spearman’s rank correlation demonstrated a significant relationship between certain Vp-1B alleles and GI. The correlation matrix contained16significant (p<0.01) correlations. GI were negatively and positively correlated with7and9Vp-IB fragments, respectively.
