| Hibiscus manihot is an annual herbage plant of malvaceous abelmosk. It contains lots ofbioactive compounds, e.g. flavonoidsã€alkaloidã€hyperinã€xanthophy1l and so on. Flavonoidshave strong antioxidant activity, and highly medicinal properties. The alkaloid has thebiological activity important component, and has the obvious pharmacology activenessvariously to the cardiovascular system, the central nervous system.In this paper,the stem and seed of Hibiscus manihot were taken as raw material, and thetraditional ethanol digestion method and the ultrasonic wave strengthening alcohol digestionmethod were adopted to extract flavonoids. The standard graph of rutin was used forcomparison, and the colorimetric technique of aluminium nitrate was employed to determinethe contents of total flavone. The extract conditions were established through the single factorexperimental method. The factors were ethanol concentration, digestion time, ultrasonic wavepower, ultrasonic wave temperature and material liquid ratio. At the same time, the bestextracting conditions was optimized through the orthogonal regression experimental designmethods. Then the flavonoids were isolated and analyzed by TLC. Finally taking rutin andquercetin as standard substance,they were isolated and analyzed by high performance liquidchromatography of ODS chromatographic column,diode array detector.Then the seed of Hibiscus manihot extracted the flavonoids and the waste liquid purifiedflavone by AB-8resin was taken as raw material for extracting alkaloid. The alkaloid wasdistinguished by the principle of in the acid solution iodizate bismith potassium reagent withthe alkaloid had responded that the production nacarat precipitates, determined the OD valueof total alkaloid by its biggest absorption wavelength. Through the single factor experimentalmethod, the optimally extract conditions were established by four factors: acetic acidconcentration, digestion time, digestion temperature and material liquid ratio. The bestextracting condition also was optimized through the orthogonal regression experimentaldesign methods. Finally the alkaloid was isolated and analyzed by TLC.The flavonoid’s optimum extracting conditions of Hibiscus Manihot seeds were70% alcohol as solvent, extracting time1.8h, extracting temperature63℃, seed material tosolvent ratio1:46.5g/mL, the extraction rate is4.03mg/g.The flavonoid’s optimumextracting conditions of Hibiscus Manihot stalks were ultrasonic wave power400W,extracting time48min, ultrasonic wave temperature57.5℃, stalk material to solvent ratio1:12g/mL, the extraction rate is1.02mg/g. The alkaloid’s optimum extracting conditions ofHibiscus Manihot seeds were0.9%acetic acid as solvent, extracted temperature37℃,extracting time60min, seed material to solvent ratio1:16.5g/mL.The study of flavonoid and alkaloid is the most active project in the world at the present.If this topic’s achievement was used into industrial production of flavonoid and alkaloid, thehuge economic interest would be brought to the enterprise, the agricultural production mayalso be driven. |
