Prolific and biologically active substances were found in the flower, stem, leaf, and seedof Hibiscus manihot. The thesis mostly refers to the research of the extraction of flavone inthe seed, flower and stem of Hibiscus manihot, as to develop the two fluorescence methodsand a HPLC method to detect the content of the flavone monomer of rutin and quercetin.In this thesis, we firstly take the seed, flower and stem of Hibiscus manihot as rawmaterial, adopt the ethanol digestion method, extract the total flavone in the Hibiscus manihot,and to make standard graph with the rutin for comparison, employ the colorimetric techniqueof aluminium nitrate to determine the contents of total flavone. Through the single factorexperimental method, we establish the extract conditions of four factors which are ethanolconcentration, digestion time, digestion temperature and liquid stalk ratio. According to thetheory of orthogonally rotational combination design, and base on the monofactorialexperiment, we take the response surface analytical method of four-factor and five-level todo experimental design, and meanwhile we get through the medium of response surfaceregression analysis means, fit the correlation of factor(ethanol concentration, digestion time,digestion temperature, aqueous stalk ratio) and response value (extraction yield) bypolynomial expression, analyse the function between the relation of factor and response valuethrough functional surface, research and optimize the correlation between the factor and theresponse value, this factor and that factor, what to do is to get the hope of obtain optimumprocessing parameter, and increase the extraction yield of flavone in Hibiscus manihot. At last,through the model of response surface and contour plots, we get the optimize processingparameter, that is when ethanol concentration is53.9%, digestion time is2.2h, digestiontemperature is76.9℃, aqueous stalk ratio is22.3mL·g-1, the extraction effect is the best andthe flavone extraction yield will be up to3.6%.Because of plants have plenty of rutin and quercetin substances, especially these twoflovonoids have been found widely application in the field of biology medical science, welargely research the rutin-potassium ferricyanide-borax system and quercetin-aluminum(Ⅲ) system in this thesis, respectively, which are primarily according to the property that thehydroxy or oxatyl of enol form were able to form PI bond conjugated system with metallicion of rutin and quercetin molecule, and form the compound in possession of the property thatsensitivity high, detect ability low, and fluorescence strong, determine the content of rutin orquercetin in Hibiscus manihot, analyze each systematic fluorescence quality and influencingfactor, get through the systematic degree of precision and coefficient of recoveryexperimentation, develop the fluorescence detection techniques about rutin and quercetin inthe plant of Hibiscus manihot.In order to make further qualitative and quantitative analysis flavonoid substances in theplant of Hibiscus manihot, we finally research and develop the isolation analysis method,which take rutin and quercetin as standard substance, with high performance liquidchromatography of ODS chromatographic column, diode array detector. The detection resultfurther declaration that the plant of Hibiscus manihot certainty maintain prolific flavonoidsubstances, and the resource are deserving of developing and spreading.
