In this dissertation, the sequential injection online dilution coupled with spectrophotometer technique was proposed and applied to the analysis of chloridion and uric acid in sample.The first section of the dissertation is the introduction of online dilution technique which was composed of online scattered dilution technology, online dialysis dilution technology and online gas diffusion dilution technology. As the two aspects of online scattered dilution technology, flow injection analysis and sequence injection online scattered technology and their application was introduced. The paper also introduced online dialysis dilution technology and online gas diffusion dilution technology and their application.The second section of the dissertation described a sequential injection online dilution coupled with spectrophotometer in determination of chloridion in milk and urine. The proposed method was based on the reaction of Cl" and Hg(SCN)2which produced micro ionization HgCl2complex and SCN" in acid condition, then the SCN" and Fe3+produced a red complex which could make a characteristic absorption. In the best experiment condition, the linear range was20-500mg/L The relative standard deviation for11repeated measurements is0.68%for300mg/Lchloridion. A throughput of15samples/h can be achieved with a recovery between92.2%and106.4%In the third section a sequential injection online dilution method was proposed for monitor uric acid in urine. The proposed method which composed of online dilution and online determination utilized online dilution technology to achieve online sample pretreatment and overcome the fault of poor precision in manual operation. The linear range was5.0×10-5-5.0×10-3mol/L and the RSD was2.2%(n=11). Compared with manual dilution, there was no significant difference.In the last section, characteristics of the experiments were summarized, and the prospect of application of sequential online dilution coupled with spectrophotometer in biological sample analysis was described. |
